Abstract


Sperm can fertilize an egg successfully requires the integrity of the plasma membrane and mitochondrial membrane potential , metabolism, maintaining the vitality of sperm capacitation, acrosome reaction and sperm-egg binding so the sperm plasma membrane integrity and mitochondrial membrane potential closely related. Sperm plasma membrane integrity and sperm mitochondrial membrane potential may be an important indicator to assess semen quality , worthy of further study . One hand , this study Rh123/PI and JC-1 staining to detect the fertile men and infertile patients with sperm mitochondrial membrane potential dual Rh123/PI staining and JC- 1 staining was evaluated in the evaluation of semen quality feasible sex . The results show that , the Rh123/PI dyeing and JC-1 staining results and sperm motility , \On the other hand , this study also explore the relationship between the sperm plasma membrane PS valgus and mitochondrial membrane potential , found that sperm mitochondrial membrane potential loss percentage and plasma membrane PS valgus percentage correlation . On the other hand , we have the semen of fertile men discontinuous density gradient centrifugation of semen dope , 45% of the layer of low vitality of sperm and 90% storey vitality of sperm containing 0.3 % human serum albumin (HSA) and 3.0 % HSA BWW culture medium incubated detected at different time points plasma membrane PS valgus , mitochondrial membrane potential and plasma membrane disorder . The results showed that 45% layer of low vitality of sperm PS valgus and mitochondrial membrane potential loss percentage than dope , 90% lower than dope ; 12h vitro incubation under different conditions , the live sperm PS valgus percentage loss of mitochondrial membrane potential percentage and plasma membrane phospholipid disorder have not seen a significant change ; the necrosis sperm increased significantly at 12h , in vitro sperm in the normal incubation process , and not spontaneous apoptosis , only to necrosis of the form into the final outcome .
