Abstract


Objective: To investigate novel tumor suppressor gene PTEN, P16 and tumor metastasis suppressor gene nm23 expression in breast cancer and clinical pathological factors and prognosis. Methods: Using immunohistochemistry SP method detected 50 cases of breast cancer surgical specimens (including lymph node metastasis in 24 cases, 26 cases without lymph node metastasis. Follow-up results of 10 patients died within five years.) And 25 cases of breast fibroadenoma primary lesions in PTEN, P16 and nm23 expression Immunohistochemical expression was observed by optical microscopy, the application of statistical methods to analyze the results. Results: 50 patients with breast cancer, of PTEN expression was 56%, the positive expression rate of P16 and nm23 were 48% and 58%, respectively. High expression of PTEN in 25 cases in the control group was 80%, the positive expression rate of P16 and nm23 were 88% and 72%. High expression of PTEN and P16 positive expression rate in the breast cancer group was significantly lower than the control group (P <0.05). nm23 positive expression rate in the two groups showed no significant difference (P> 0.05). In breast cancer, the high expression of PTEN protein, P16 and nm23 positive rate in histological grade among the three groups, a significant difference (P <0.01); of PTEN expression and nm23 positive rate in clinical stage among the three groups a significant difference (P <0.01); positive rate of P16 and nm23 between lymph node metastasis were significant differences (P <0.05). High expression rate of PTEN, nm23 and P16 positive expression rate of survival time ≥ 5 years and <5 years between the two groups, a significant difference (P <0.05). COX regression model analysis of histological grade, axillary lymph node metastasis of PTEN and nm23 four indicators reflect the prognosis of breast cancer risk for 3.331,4.706,0.101 and 0.179, respectively, the PTEN high expression and nm23 positive expression when the risk is small. Conclusion: The expression of PTEN and P16 in breast cancer is significantly lower than that of breast fibroadenoma. P16 and nm23 and breast cancer cell differentiation, lymph node metastasis of. PTEN and nm23 there is a protective effect in breast cancer tumor progression, and prolong the survival time of the patients. The positive expression of PTEN and nm23 is indeed protective factors, and have the opportunity to increase the survival time of patients. Therefore, PTEN, P16 and nm23 determine the activity of breast tumor cell proliferation, invasion and metastasis important indicators, combined detection of breast tissue PTEN, P16 and nm23 expression status can help determine prognosis.
