Abstract


The purpose of detection of different concentrations of amlodipine gel transdermal ability to filter out the cumulative permeation amount to achieve a lower minimum effective amount of amlodipine concentration . Observe the the Amlodipine gel concentration of rat ischemic random flap survival and to detect flap Amlodipine retention amount , to explore the relationship between the two . Method of in vitro transdermal experiments using modified Franz diffusion cell , and after the administration of different times of UV spectrophotometric breadth assay to accept pool amlodipine cumulative penetration amount , screening out the most have to promote the penetration effect of penetration enhancers and cumulative permeation amount larger minimum amlodipine concentration . Establish dorsal ischemic random flap pedicle in the end of the model (10mm × 80mm), immediately after starting the flap surfaces were coated with 05% , 1.5% amlodipine gel and ammonia amlodipine , condensate plastic matrix 2g once daily for seven consecutive days , 7 days measured coordinate paper flap survival area ; addition, in the first 2 and 6h after administration along its longitudinal axis , respectively, cut 1 g of the total length of the flap tissue , homogenized , after extraction detecting amlodipine retention amount. Results 1. In vitro transdermal experiment , 0.5% , 1.0% amlodipine gel cumulative permeation amount is less than 1.5% , 2.0% , 2.5% amlodipine gel ( P <0.05 ) , 0.5 % and 1.0% ammonia amlodipine gel and 1.5% , 2.0% and 2.5% of amlodipine gel no difference (P> 0.05), but the different concentrations of amlodipine gel cumulative penetration increases with the use of time and increased ( P < 0.05). 2.0.5 % amlodipine the gel group of the flap survival area of ??391.4 ± 65.4 mm ~ 2, higher than the 192.9 ± 56.8 mm to 2 ( P <0.05 ) , although 1.5% of amlodipine gel also improve flap survival area , but no significant difference compared with the control group ( P = 0.217 ) . 3. The flap the amlodipine lifetime amount of 0.5% in the amlodipine group was significantly lower than the 1.5% gel the Amlodipine gel group ( P <0.05 ) . Conclusion 1. Amlodipine percutaneously cumulative penetration amount increases with the drug concentration and the increase of the penetration time . 2. Comparative the Amlodipine gel with 1.5% , 0.5% amlodipine gel can significantly improve the ischemic skin flap survival area .
