Abstract


Objective: To ivestigate the inhibitory effects of celecoxib, Toremifene and xihuangwan single use or combination on the occurrence and proliferation of experimental mammary carcinoma. Content: Sprague-Dawly rats were divided randomly into six groups : DMBA , DMBA with celecoxib, DMBA with Toremifene, DMBA with xihuangwan, DMBA with celecoxib and xihuangwan, DMBA with Toremifene and xihuangwan.At 120 days after DMBA adminstration,all rats were killed to be observed the incidence of breast cancer, Samples of breasts were collected for light microscope observation . Methods: DMBA induced mammary carcinoma was produced in rats. Chemicals had been intragastric administered to group A , group B , group C , group D , group E and group D respectively for 120 days. Results:【1】Mammary carcinoma incidence of group B, group C, groupE, groupF was significantly lower than group A (B: A degrade 45 %, C:A degrade 50%, E:A degrade 75 % ,F:A degrade 70%, P<0.05) ;tumor load(B:A degrade 57%, C:A degrade 55%, E:A degrade 87.5%,F:A degrade 84.3%, P<0.05) ;tumor diameter (B:A degrade 36.8%, C:A degrade 40.5 %, E:A degrade 65.7%,F:A degrade 62.1%, P<0.05)【2】Mammary carcinoma incidence , tumor load , tumor diameter of group B was significantly higher than group E (↑30%、29.6%、28.9%, P<0.05); Mammary carcinoma incidence , tumor load , tumor diameter of group C was significantly higher than group F (↑20 %、29.6%、21.5%, P < 0.05); Mammary carcinoma incidence , tumor load , tumor diameter of group D was significantly higher than group F (60%、60.9%、54.7%, P<0.05); Mammary carcinoma incidence , tumor load , tumor diameter of group D was significantly higher than group E (65%、57.8%、51%, P<0.05);median detection (50% of tumors)of group A occurred at 70 days after DMBA induction compared with 88 days(groupB) and 90 days(groupC) and 102days(group E) and 105 days(group F) in the treatment groups, respectively. (P<0.01). Conclusion:These results help confirm the chemopreventive activity of celecoxib, Toremifene and xihuangwan single use or combination against breast cancer of SD rats. The combination possesses strongger chemopreventive activity than single use. It illustrates that the combination of Chinese crude drug and western medicine have the best chemopreventive activity.
