Objective To investigate aminoguanidine (Ag) and resveratrol (Res) on oxidative stress in diabetes mellitus (DM) in rat liver and the protective effect of diabetes liver disease. Methods March 50, age healthy male Wistar rats were fed a high fat and sugar diet, weight about 400g, randomly divided into 5 groups. Normal control group (NC group) 8, the remaining 42 streptozotocin (STZ, 35mg · kg -1 sup>) by intraperitoneal injection to establish a model of diabetes. Modeling, one week after the diabetic rats were divided into diabetic control group (DM group, n = 10), aminoguanidine (50mg · kg -1 sup> · d -1 sup>) treatment group (Ag group, n = 10), resveratrol (10mg · kg -1 sup> · d -1 sup>) treatment group (Res group, n = 11) and combination therapy (50mg · kg -1 sup> · d -1 sup> Ag 10mg · kg -1 sup> · d -1 sup> Res) group (Ag Res group, n = 11). NC group and DM group was given the same amount of solvent intraperitoneal injection, the rats were sacrificed after 9 weeks of treatment with phlebotomy. Using colorimetric determination of malonic aldehyde (MDA) in rat liver homogenate and liver mitochondria, Cu / Zn superoxide dismutase (Cu / Zn-SOD) activity and total antioxidant capacity (T-AOC). Whole blood fasting blood glucose and serum alanine amino transfer enzyme (ALT), aspartate aminotransferase the enzymatic (AST), triglyceride (TG) and total cholesterol (TC). Using colorimetric determination of the serum hydroxyproline content and liver enzyme CYP450 2E1. Using light microscopy and transmission electron microscopy observation of liver tissue pathological changes and ultrastructural changes. Results 1. Compared with NC group, the DM rats liver homogenate and liver mitochondria MDA content was significantly higher (P <0.01), and Cu / Zn-SOD activity, T-AOC significantly lower (P <0.01); and DM groups compared to Ag group, Res Group and Ag Res group could significantly reduce liver homogenates and liver mitochondrial MDA content (P <0.01), the Res group of liver homogenate Cu / Zn-SOD was significantly higher (P <0.05), but still lower than the NC group, the of Ag Res group of Cu / Zn-SOD increased more significantly, and close to the NC group (P> 0.05), Ag group, Res Group and Ag Res group can improve liver homogenates T-AOC ( P <0.05) and liver T-AOC (P <0.01). 2. Fasting glucose levels in diabetic rats was significantly higher than the NC group (P <0.01), Ag group, Res Group and Ag Res group had no significant decrease of blood sugar. 3. DM rats liver damage and fat metabolism, Ag and Res protective role, in particular, to reduce the role of serum ALT combination therapy appears to reduce the role of the AST; Res reduce serum TG was more effective. 4. Compared with DM group, Ag group, Res Group and Ag Res group could reduce the the serum hydroxyproline content, can also reduce the activity of liver mitochondrial CYP450 2E1. 5. Three treatment groups liver of diabetic rats have a certain degree of protection in the light microscope and transmission electron microscope level. Conclusion 1. Diabetic rats obvious liver lesions, oxidative stress may be involved in the occurrence of diabetic liver disease. 2. Ag and Res reduce the activity of the liver mitochondrial CYP450 2E1 reduce liver mitochondrial lipid peroxidation, improve diabetes liver oxidant / antioxidant imbalance, has a protective effect on diabetic liver disease. 3. Ag and Res diabetes liver disease may have a synergistic therapeutic effect.
