Abstract


High frequency regeneration system of filtering the resulting salt-tolerant strains of alfalfa (Medicago sativa L.) varieties and normal callus growth resulting plants to high temperature and humidity , \0% , 0.2% , 0.4% , 0.6% , 0.8% and 1.0 % NaCl solution stress malonic aldehyde ( MDA ) , proline (PRO), soluble protein (SP) and soluble sugar content (SS), and superoxide dismutase (SOD) activity , physiological and biochemical indices of quantitative analysis and comprehensive evaluation of the salt tolerance of salt-tolerant mutant lines were identified . Agrobacterium-mediated transformation of BADH ( betaine aldehyde dehydrogenase ) gene , to establish a set of Agrobacterium -mediated the alfalfa gene conversion optimization system . Saline-alkali alfalfa strains bred for saline land development plant germplasm information . The main results are as follows : 1 ) alfalfa salt-tolerant mutant material malondialdehyde , proline , soluble protein and soluble sugar content , and SOD activity were significantly (p lt; 0.05) or highly significant with ( p lt ; 0.01) higher than unfiltered control plants . 2 ) with the NaCl concentration increased , proline and soluble protein content of salt-tolerant variant line alfalfa materials showed an increasing change of malondialdehyde (MDA), showed a down then up change ; soluble sugar content and SOD activity changes increased first and then decreased , more salt concentration of 0.8% as a turning point in the curve . 3) the salt tolerance Composite of salt-tolerant variant line alfalfa varieties : the \4) to cultivate of 7d Gannon on the 4th and prosperity alfalfa aseptic seedling hypocotyl explants , Agrobacterium strain mediator , has been optimized the alfalfa gene conversion system . Cefotaxime 200mg / L , the inhibition of Agrobacterium better ; the Kan screening concentration : Gannon Golden Age 40mg / L, 50 mg / L 4 . Hypocotyls test object ; broth OD600 of 0.3-0.5 , infection time 10min ; , get 10 anti- Kan Gannon transformed plants were screened , including a positive by PCR test . It showed BADH gene in transgenic plants with good expression .
