Abstract


Purpose Hedysari separation and extraction of HPS , and their levels were measured . Investigate the the red HPS on human gastric cancer MGC-803 cells inhibition induced tumor cell apoptosis . The method of water extraction and alcohol precipitation method the crude , sevag deproteinized , ethanol fractionation , Sephadex G - 200 column chromatography purification of HPS , phenol - sulfuric acid spectrophotometric determination of polysaccharide content . Of HPS on the inhibition of the MGC-803 cells by MTT assay , fluorescence microscopy, electron microscopy , flow cytometry technique to observe and detect apoptosis . 1: The extraction and purification of single uniform polysaccharides , the average results of the polysaccharide content of 94.88 % , RSD = 1.05 % . 2:1.0-4.0 mg / ml of red HPS on the MGC-803 cells in vitro significantly inhibited compared with the control group, a significant difference ( P <0.05 ) inhibition with the role of time and polysaccharide concentration increases while the increase in a time and dose dependent . 3 : Fluorescence microscopy apoptotic cells showed dense nuclei were stained blue nucleus color some white , some nuclear staining solid shrunk the high agglutination punctate structures , the formation of apoptotic bodies . Under transmission electron microscopy for the cells to the apoptotic cells showed pyknotic nuclear chromatin , and formation of crescent -shaped body , cytoplasmic vacuolization , and formation of apoptotic bodies . 4 : flow cytometry Sub-G 1 apoptosis peak G 1 before , G 0 / G 1 phase cells increased , compared with the control group, a significant difference ( P <0.05 ) . Conclusion 1 : Red HPS MGC-803 cells cultured in vitro growth inhibition may prevent cell cycle progression and induce tumor cell apoptosis . Its role in blocking cell cycle in G 0 / G 1 of related . 2 : column chromatography separation polysaccharides can make up for the low insufficient ethanol precipitation accuracy phenol - sulfuric acid spectrophotometric method is simple , accurate , the red HPS Determination get satisfactory results .
