Abstract


Cytoplasmic male sterility (CMS) is an important way to make use of rapeseed heterosis . The the Brassica napus polima cytoplasmic male sterility (pol CMS) CMS types of Brassica napus , is widely used in the three-line or two-line hybrid breeding at home and abroad . At present , domestic and foreign pol CMS has bred a large number of excellent hybrids , and large scale application . Studies have shown that : the pol CMS sporophyte infertility type, infertility the plasmon atp6/orf224 the expression restore gene nucleus . In view of the nuclear restorer genes play an indispensable role in the the pol CMS three-line or two-line heterosis utilization in cloning recovery has great significance in the recovery mechanism of the gene in breeding applications as well as male sterility . This study successive backcross build the pol CMS restorer gene nearly isogenic line population for the materials , screening and recovery molecular markers tightly linked to the gene (Rfp) , and its initial positioning of the map , in order to further fine mapping and cloning of the foundation . The main results obtained are as follows : 1 . Choose fertile genotypes monoclonal S (Rfprfp) the emasculation and recurrent parent the N (rfprfp) of successive backcrossing containing 980 fertile plants , BC 12 1044 sterile plants F 1 groups (χ2 test is consistent with the theory of 1:1 segregation ratio ) , was successfully constructed carrying pol CMS Restorer Gene nearly isogenic lines . 2 segregating population Group hybrid analysis method ( BSA ) , screening of 3840 pairs of AFLP primer get 15 Rfp tightly linked AFLP markers , these markers located on the same side of the Rfp gene marker , and Rfp genetic distance for the nearest 0.65cM . Four AFLP markers of the the Rfp genetic closest was successfully converted into the SCAR the mark (SCCQ1 SCCQ4 ) , which tag SCCQ1 , SCCQ3 and validate the different sources of recovery materials SCCQ4 found better versatility , can be applied restorer lines of molecular marker-assisted breeding and laboratory recovery rate of identification of the hybrid configuration . 4 4 SCAR marker sequence of Arabidopsis sequence comparison and integrated development of our group the SSR markers SSRkbr ( , Rfp distance 0.8cM) integrate information linkage map with TN , will restore gene Rfp positioning in Arabidopsis mustard on chromosome 1 .
