Glycocholate ( GCA ) is one of the main active ingredient of cattle and sheep bile . This study was designed to investigate the apoptosis in vitro and in vivo in the GCA role of mouse spleen lymphocytes . Inverted microscope was used to observe the nuclear morphology of spleen lymphocytes , spleen lymphocyte DNA fragments by agarose gel electrophoresis , detected by flow cytometry of spleen lymphocyte apoptosis early phosphatidylserine percentage . Spectrophotometric Determination of apoptotic cells redox reactivity - GSH content of Caspase-3 and Caspase-9 activity was detected by the ELISA method . The in vitro results showed that : GCA spleen lymphocytes produce nuclear condensation and apoptotic bodies ; significant cause spleen lymphocyte DNA fragmentation ( P lt ; 0.05 ) significantly induced spleen lymphocyte phosphatidylserine ( P lt ; 0.01 ) ; significantly reduced GSH content ( P lt ; 0.05 ) ; significantly increased Caspase-9 and Caspase-3 activity ( P lt ; 0.05 ) ; vivo results showed that : the GCA can make spleen lymphocytes to produce nuclear condensation and withered death corpuscles ; significant cause spleen lymphocyte DNA fragment of ( P lt ; 0.05 ) ; able to extremely significantly increase the percentage of phosphatidylserine valgus ( P lt ; 0.01 ) ; significantly reduced GSH content ( P lt ; 0.05 ) ; significantly improve caspase - 9 and caspase - 3 activation level ( P lt ; 0.01 ) Conclusion : GCA on the body , outside the spleen lymphocyte apoptosis could promote the role .
