Objective To study the keratin 19 (Cytokeratin 19 or Keratin 19, CK19) expression in NPC tissues and cells, and to analyze the relationship between its expression and nasopharyngeal carcinoma clinicopathological features. Methods 62 patients with nasopharyngeal carcinoma, 17 cases of metastatic nasopharyngeal carcinoma and 16 cases of chronic nasopharyngitis (control group) as a normal nasopharyngeal mucosal specimens by pathologically confirmed by immunohistochemical SP method detected CK19 expression levels, and analysis of expression levels with clinical and pathological features of nasopharyngeal; different degrees of differentiation and with different metastatic potentials detected by western blot CK19 protein expression in NPC cell lines. 1) immunohistochemical results show CK19 expression in the cell cytoplasm and cell membrane, nasopharyngeal CK19 positive rate of 74.2% (46/62); positive metastatic nasopharyngeal carcinoma was 88.2% (15/17 ); control group of normal nasopharyngeal epithelial positive rate was 12.5% ??(2/16); of CK19 expression in lymph node metastatic nasopharyngeal carcinoma than nasopharyngeal specimens in the NPC is higher than the normal control group, there were significant differences significance (P lt; 0.05) 2) CK19 expression in NPC differentiation related to: CK19 positive rate was 46.2% (6/13) in the moderately differentiated nasopharyngeal carcinoma, poorly differentiated nasopharyngeal positive rate was 81.6% (40/49 ), the difference between the two was significant (P lt; 0.05). 3) CK19 expression NPC primary tumor size: CK19 expression in non-intrusive group the positive rate was 20.0% (1/5), while in the surrounding violations of the group, the positive rate was 78.9% (45/57), two between There was significant difference (P LT; 0.05). 4) CK19 expression with NPC lymph node metastasis: CK19 expression in the absence of lymph node metastasis of NPC in the positive rate was 11.1% (1/9), NPC lymph node metastasis was 84.9% (45/53) between the There was a significant difference. 5) CK19 expression and NPC distant knot metastasis: CK19-positive NPC without distant metastasis rate was 71.4% (40/56), 100% (6/6 in distant metastasis NPC), the two between There was significant difference (P LT; 0.05). 6) of CK19 protein expression and NPC clinical staging: I II period NPC tissue expression of CK19 positive rate was 45.5% (5/11), Ⅲ Ⅳ period NPC in CK19 expression positive rate was 78.8% (41 / 52), between the two there are significant differences (P LT; 0.05). 7) of CK19 protein expression and gender, age has nothing to do (P gt; 0.05) 8) Western blot analysis results showed that the level of expression of CK19 in poorly differentiated NPC cell line CNE2 the higher differentiation NPC cell lines CNE1 significantly raised; highly metastatic NPC cells Department of the lower level of expression in 5-8F transfer of NPC cell line 6-10B were significantly increased (P lt; 0.01), CK19 expression NPC differentiation and metastasis. Conclusion 1) CK19 expression than the primary tumor in metastatic nasopharyngeal chronic nasopharyngitis, higher than in NPC; nasopharyngeal carcinoma poorly differentiated and highly metastatic nasopharyngeal carcinoma cell CK19 expression above good differentiation and low metastatic potential of nasopharyngeal carcinoma cell line, CK19 as nasopharyngeal cancer differentiation and metastasis molecular markers. 2) of CK19 expression and primary tumor size, degree of differentiation, lymph node metastasis and distant metastasis, clinical stage. Age, regardless of gender.
