Objective: diallyl disulfide (diallyl disulfide, DADS) induced CNE2 cells apoptosis-related genes , to explore the molecular mechanisms of DADS -induced apoptosis of CNE2 cells using gene chip analysis . Methods: MTT assay DADS inhibition of the CNE2 cell proliferation , the IC50 value ; Cell growth curve analysis DADS CNE2 cell growth inhibition ; Hochest staining , flow cytometry and DNA gel electrophoresis to detect CNE2 cell apoptosis induced by DADS the case ; gene chip , DADS induced CNE2 cell apoptosis-related genes ; detected by RT- PCR validation of gene and protein expression and Wester blot . Results: 1 DADS inhibit the proliferation of CNE2 cells ICso value of 30 mg · L-1; With DADS concentration increases , the processing time extension of CNE2 cells to inhibit significantly increased ( P lt; 0.05 ) . 2, Hochest staining showed typical apoptotic morphological changes induced CNE2 cells with 30mg · L-1 DADS . Flow cytometry showed CNE2 cells apoptosis rate increased gradually with increasing ] DADS concentration ( P lt; 0.05 ) . 4 , agarose gel electrophoresis showed that , with the DADS concentration reached 30mg · L-1 , showing the DNA ladder . 5 , the gene chips were detected 1074 differentially expressed genes , including apoptosis related gene 24 ( 19 up-regulated genes downregulated genes , 5 ) . 6, RT-PCR and Wester-blot results confirmed the upregulated genes SON and down the gene IL24 gene microarray results . Conclusion: 1 , DADS can induce apoptosis in CNE2 cells 2 DADS - induced CNE2 cell apoptosis may be of of TRAF1, CASP9, FOXO1A and IL24 and other genes upregulated and PIK3R3 ELMO1 , SON and DFFB , , such as gene expression downregulation .
