Abstract


Ovarian cancer is a common gynecological cancer mortality rate ranks first in the Gynecologic Oncology. Advanced ovarian cancer five-year survival rate of only 20% -30%. Ovarian cancer occult onset, symptoms only in the spread of the disease to the peritoneal cavity surface when manifested, it is difficult in the early detection and diagnosis. However, during this period, has not by surgical removal of all lesions, leading to relapse after surgery. Therefore, the main treatment options for advanced ovarian cancer chemotherapy, ovarian cancer usually was progress in the state and easy to chemotherapeutic drugs tolerated. Therefore, further studies of the pathogenesis of ovarian cancer, and to find new effective methods of treatment of ovarian cancer is very important for prevention of recurrence after treatment and surgery. MicroRNAs discovered in recent years the regulation of gene expression at the transcriptional level, length RNA molecules about 18-24, and the regulation of gene expression through mRNA-3'UTR region of the target gene pairs. It has been proved that miRNAs can regulate a variety of physiological and pathological processes, such as cell differentiation, cell proliferation and tumor formation. The miRNAs can be used as oncogenes or tumor suppressor gene, a plurality of signal paths pathway involved in the regulation of human tumor formation. Scholars use miRNA expression microarray variety of methods to detect ovarian tissue of ovarian tumors and normal female miRNAs expression found abnormal expression of ovarian cancer, miR-100 and other miRNAs, but the mechanism of the specific role of miRNAs in ovarian cancer not yet clear. We found nine cases of ovarian serous carcinoma and four cases of normal ovarian tissue miR-100 expression by RT-PCR analysis, compared with normal ovarian tissue, miR-100 were down-regulated in ovarian serous carcinoma. At the same time, we found that overexpression of miR-100 can reduce ovarian cancer cells to cisplatin resistance in ovarian cancer cell lines, but does not affect the growth of ovarian cancer cells; through animal experiments using nude mice in vivo analysis of tumor neovascularization generation, found that over-expression of miR-100 inhibit ovarian cancer angiogenesis. In order to clarify the mechanism of action of miR-100 in ovarian cancer, we luciferase reporter gene that miR-100 directly targeting mTOR-3'UTR and inhibit its expression. We speculate that miR-100 on mTOR regulation may have an important role in ovarian cancer formation and angiogenesis. The results of this study will help to understand the role of miR-100 in ovarian cancer, and to provide a theoretical basis for the prevention and treatment of ovarian cancer.
