Abstract


Antimicrobial peptides (antibacteral peptides, ABP), a class of biologically active small peptides widespread in the biosphere , generally have anti- bacterial or fungal , and some also have the function of the the antiprotozoal virus or tumor cells without toxic side effects , times favored by domestic and foreign scholars , has become a research focus of domestic and foreign scholars . Chinese forest frog amphibians belong to the land and sea life , the exposed skin , long-term process of natural selection , and gradually formed a series of protection mechanisms , in order to resist the invasion of harmful environmental factors . Wherein, in the to microbicidal active peptide containing a large number of their skin . In order to verify Chinese forest frog skin antimicrobial active substances , fresh Rana skin as experimental material , with a 80% methanol solution extraction via SephadexG-100, SephadexG-50 and SephadexG-25 gel purification , to obtain high purity the antimicrobial active substance. The purified antimicrobial substances by Escherichia coli, Staphylococcus aureus, Bacillus subtilis , Pseudomonas aeruginosa , yeast do bacteriostatic test , the minimum inhibitory concentrations were 6.37μmol . L -1 , 3.18μmol. L -1 , 6.37μmol. L -1 , 6.37μmol. L -1 , 25.48μmol. L -1 . Forest frog skin antimicrobial active substances purified by urea -SDS-PAGE electrophoresis analysis , performance is the one with a molecular weight of approximately 6.28KDa . Antibacterial substances in the purified Rana skin by cellulose acetate membrane electrophoresis , showed that having a strong cationic character ; by amino acid analysis , indicating that the Rana skin antimicrobial substances from alanine , glycine , leucine, isoleucine acid , proline , Isolation , purification and identification of Chinese forest frog skin antimicrobial peptides is a summary of glutamic acid , threonine , serine , lysine . China Rana skin containing the antimicrobial active substance is a small stars in the peptides , i.e. the antibacterial peptide . Isolation , purification and identification of Chinese forest frog skin antimicrobial peptides , laid a certain foundation to explore biologically active substances the antimicrobial peptides medicinal development value .
