The Effect of Intra-articular Injection with Doxycycline on the Early Degeneration of Opposite Knee of the Immobilizied Rabbit Model Osteoarthritis

Abstract


Objective: To study the side of intra-articular injection of doxycycline on the opposite side of the brake rabbit osteoarthritis model the effect and mechanism of early degeneration of the knee . Methods: New Zealand white rabbits were 36 weight , gender randomly divided into blank control group , model control group and the experimental group , the control group 6 , model control group , 18 in the experimental group 12 . Model control group 4,8,12 W three different time points were randomly divided into 3 groups of 6 . Experimental group were randomly divided into 2 groups of 8,12 W two different points in time . Model control group and the experimental group were treated with cast immobilization left lower limb . Plaster for 4 weeks after the daily administration group given 0.3 ml of 1.33% doxycycline left knee joint intraarticular injection , and then sacrificed rabbit taken the detection of the right knee joint cartilage associated indicators . Results : (1) compared with the blank control group , model group cartilage surface darken the cartilage surface ulceration mankin score was significantly higher than that of the control group ( P lt ; 0.05) , blood and joints of MMPs -3 , of IL- higher 1β expression (P lt; 0.05). Synovial fluid of iNOS and NO expression higher ( P lt ; 0.05 ) ( 2 ) compared with the model group , treatment group cartilage surface is smooth , mankin scores (P lt; 0.05), blood and joints in of MMPs - 3 IL-1β expression lower (P lt; 0.05). Synovial fluid expression of iNOS and NO lower (P lt; 0.05), Conclusion: (1 ) brake rabbit osteoarthritis model contralateral knee due to an increase in weight-bearing , biomechanical balance changes and related cytokines and media through blood also be the performance of early degeneration of articular cartilage , and over time , the degree of degeneration and heavier . (2) after intra-articular injection of doxycycline by local effects and blood role on the opposite side of the rabbit knee osteoarthritis cartilage early degeneration significantly alleviate the effect from the effective prevention and treatment of osteoarthritis inflammation early lesions . ( 3) doxycycline early articular degeneration have eased significantly the role of one of its mechanism of action is through the cartilage, synovial , blood , synovial fluid in the expression of MMP -3 , IL -1β , iNOS and NO expression suppression to completed.
