Abstract


The purpose of: (1) investigate the 5,10 - methylenetetrahydrofolate reductase (MTHFR) gene C677T locus mutation distribution mutation distribution with congenital heart disease (CHD). (2) the understanding of the distribution and their children of the mother (MTHFR) gene C677T gene locus mutation locus mutation associated. (3) to understand the relationship between environmental factors with CHD and to identify its risk factors, and to explore effective ways of working and modes of prevention for the future prevention of CHD. Methods: (1) survey of 100 CHD patients and 200 control subjects, in accordance with the 1:2 matched case-control study (based on gender, age, place of residence to match cases by clinical and ultrasonography for diagnosis). By polymerase chain reaction - restriction fragment length polymorphism (polymerase chain reaction restricted fragment length polymorphism, PCR-RFLP) and digestion of children and their mothers MTHFR C677T genotype distribution and its difference, explore the MTHFR C677T mutation whether related with CHD the CT allele will increase children's risk of CHD, while investigating the parents during pregnancy and pre-pregnancy exposure to environmental factors. (2) design the Child Care born quality influencing factors questionnaire survey interview CHD children and control children's parents to get the information if they are exposed to environmental risk factors. (3) single factor conditional logistic regression analysis of 28 possible risk factors for CHD, screening 16 and single factor conditional logistic regression analysis, the risk factors associated with CHD Multivariate conditional logistic regression analysis. And simultaneously to factor analysis, in accordance with the size of the factor of contribution rate, to identify a common factor. Results: (1) in CHD case group MTHFR gene 677C → T mutation control group; (2) mother MTHFR gene 677TT offspring of the occurrence of CHD have some relevance. (3) Multivariate conditional logistic regression analysis of risk factors that can be obtained by: pregnancy-induced hypertension, antipyretic analgesics, pesticides, early pregnancy, dietary and vegetable intake, negative life events, alcohol consumption, children's genotype, the OR The values: 29.76,10.194,15.156,0.414,4.69,6.302,2.333,3.474. (4) Factor analysis can be a common factor: the dietary nutritional factors, negative life events and factors, flu-related factors, alcohol and tobacco factor, miscarriage related factor, pesticides and vegetables factors, pregnancy complications factor gene factor. Conclusions: (1) mother MTHFR gene have some association with offspring CHD. (2) CHD patients MTHFR gene 677C → T mutation and the occurrence of CHD association. (3) certain environmental factors and the occurrence of CHD, by improving the environmental factors, may achieve the purpose of prevention of CHD.
