Objective: To investigate different doses of recombinant human erythropoietin (recombinant human erythropoietin rHu-EPO) blood flow dynamics of rats with myocardial infarction, left ventricular infarct size, myocardial apoptosis and proapoptotic protein mitochondrial fusion fibroin 2 (mitofusin 2, Mfn2) expression. Methods: 128 adult SD rats were randomly divided into myocardial infarction group rHu-EPO high-dose treatment group rHu-EPO (loading dose of 5000 IU / kg large doses of rHu-EPO 1500 IU / kg maintenance therapy one week), rHu group of the-EPO small dose treatment rHu-EPO (loading dose of 5000 IU / kg small doses of rHu-EPO 750 IU / kg maintenance therapy one week) and sham group, n = 32. 24 hours and two weeks using carotid intubation detection hemodynamic indicators were TTC staining left ventricular infarct size, terminal deoxynucleotidyl transferase-mediated nick end labeling (TUNEL) detection of myocardial cells apoptosis, immunohistochemical method detection Mfn2 protein expression. Results: After 24 hours: Compared with the sham group, the control group of myocardial infarction left ventricular systolic pressure (LVSP), left ventricular end-diastolic pressure (LVEDP) and left ventricular pressure rise and fall rate (± dp / dtmax) was significantly deterioration (P lt; 0.05); compared with myocardial infarction in the control group, large, small dose group ± dp / dtmax there are significant with improvement (P lt; 0.05); each surgical group myocardial infarction area without significantly with differences (P gt; 0.05 ); compared with the sham group, the apoptotic index of myocardial infarction and Mfn2 protein expression was significantly increased (P lt; 0.05); significantly reduced compared with the MI group, the apoptosis index of small-dose treatment group and Mfn2 protein (P lt; 0.05); apoptotic index of high-dose treatment group and low dose treatment group and Mfn2 protein expression was no significant difference (P gt; 0.05). After 2 weeks: sham group compared to the control group of myocardial infarction LVSP, LVEDP and ± dp / dtmax significantly worse (P lt; 0.05); compared to the control group with myocardial infarction, a large, low-dose treatment group ± dp / dtmax have significantly improved (P lt; 0.05), infarct size was significantly reduced (P lt; 0.05); compared with the large dose, small dose group ± dp / dtmax have significantly improved; compared with the sham group, myocardial apoptotic index infarction and Mfn2 protein expression was significantly increased (P lt; 0.05); compared with the MI group, the apoptotic index of small dose group and Mfn2 protein expression significantly reduced (P lt; 0.05); big dose treatment The apoptotic index of the group with low-dose treatment group and Mfn2 protein expression was no significant difference (P gt; 0.05). Conclusion: immediately after myocardial infarction loading dose of rHu-EPO and postoperative 1 week of treatment can be significantly reduced myocardial infarction in rats infarct size reduction to maintain, protect heart function, and reduced to promote the expression of apoptotic protein Mfn2 reduce myocardial apoptosis. Compared with the high-dose group, the effect of low-dose group to improve heart function better, but the low-dose group remained administration against myocardial apoptosis and regulation of Mfn2 protein were not significantly different.
