Abstract


Filariasis is a serious harm to human health, infectious diseases , long - term use of chemical drugs , filarial and mosquito-borne resistance enhanced . Research and development of low-cost , safe and efficient filarial vaccine is an important issue filariasis work potential . DNA vaccine developed in recent years for its preparation is simple , practical and efficient and much attention has broad application prospects . Available data indicate that the parasitic worms and protozoa , 3 - GAPDH (glyceraldehyde-3-phosphate dehydrogenase, GAPDH) can be induced in experimental animals to produce a protective immune response , is an important vaccine candidate antigen . This study successfully constructed the cycle type malayi DNA vaccine pcDNA3.1/Bm-gapdh . In order to improve the immune effect , immunostimulatory the sequence CpG as adjuvant co- immunized BALB / c mice , using RT-PCR, MTT, ELISA detection of GAPDH immune effector . The results indicate that the anti- filariasis DNA vaccine candidate molecule nucleic acid the vaccine pcDNA3.1/Bm-gapdh has the potential to increase the adjuvant immune effect has been enhanced . In order the next vaccine designed for the preparation , the experiment also constructed periodic Brugia malayi cystatin (cysteine ??protease inhibitor, CPI) recombinant plasmid pGEM-T/Bm-cpi in and sent to a gene Company sequencing. Then , according to the sequencing results , suggesting that the amino acid sequence , and then using the relevant software to predict its B cell epitope .
