The Protective Effect of Acanthopanax Prescription on Myocardial Ischemia Reperfusion Injury in Rabbits and Its Mechanism

Abstract


Objective: This study myocardial ischemia-reperfusion injury in rabbits as a model of Acanthopanax square myocardial ischemia and reperfusion injury protective effect and mechanism of action, and to provide a scientific basis for the development of new drugs. Methods: 24 Japanese white rabbits were randomly divided into three groups: the sham group (A), model group (B), the the Acanthopanax party group (C group). Animals in each group were normal saline and drug orally daily 10ml/kg. Seven weeks after the last gavage 1h underwent surgery. The animals ear vein injection of 25% urethane (4ml/kg) anesthesia back bit fixed neck midline incision endotracheal intubation. The the thoracic midline incision exposed heart, pierced the pericardium, clamping the left atrial appendage gently on mentioning valgus below the left atrial appendage roots of 2-3mm wear silk suture anterior descending coronary artery (A group only wear lines without ligation) loosen the ligature after 30min, coronary perfusion recanalization 3 h. In each experimental group were perfusion in 3h arterial blood measured hemorheology; blood rapidly after the animals were killed approximately 100 mg taken ischemic myocardial tissue made from tissue homogenates using automatic biochemical analyzer SOD, GSH- Px activities, MDA content changes; immunohistochemistry SABC method groups myocardial tissue expression of Bcl-2 and Bax. ECG machine records ligation ligation after 5min, 20min, 40min and reperfusion for 1h, 3h II-lead ECG measured compare before and after ischemic ST-segment changes each time period. The result: a model group compared with the sham group, MDA increased significantly; the Acanthopanax party group the MDA significantly lower than the model group, the difference was significant. Model group compared with the sham group, SOD, GSH-Px activity decreased significantly, there are significant differences; the Acanthopanax party group can significantly increase serum SOD and GSH-Px activity, compared with model group was significantly different. 2 model of NO and NOS activity was significantly lower than the sham group, and Acanthopanax party group Acanthopanax side can significantly increase the myocardial tissue NO production and NOS activity. 3 model group compared with the sham group, whole blood, high shear viscosity, plasma viscosity, etc. are very significant differences, indicating that the models were successfully. The Acanthopanax party group can significantly reduce acute myocardial ischemia-reperfusion injury status rabbits low cut and high shear viscosity of whole blood, plasma viscosity compared with model group with a significant difference. Bcl-2 content higher than 4 the Acanthopanax square myocardial tissue in the model group, Bax content lower than the model group, the difference was statistically significant, whereas no difference to the levels of Bcl-2 and Bax in the myocardial tissue of sham group . Conclusion: Acanthopanax side significantly reduce blood viscosity index improve sluggish, viscous blood flow, improve myocardial microcirculation; standard II lead ECG ST-segment downward trend, thereby protecting ischemia reperfusion heart. 2 Acanthopanax recipe could reduce ischemia-reperfusion myocardial MDA content, increased NO, SOD, GSH-PX and NOS activity, suggesting a significantly scavenge oxygen free radicals to protect myocardial tissue. 3 the Acanthopanax party to promote myocardial tissue of Bcl-2 expression, inhibition of the expression of Bax, play inhibit myocardial cell apoptosis, indicating Acanthopanax the myocardial protection role and regulation of Bcl-2 and Bax expression.
