Abstract


Objective: factor analysis of inflammatory factors and Th17 cells in the pathogenesis of asthma and, to explore the of Uygur medicine God vanilla inflammation in asthma and Th17 cells related factors, to clarify the treatment of asthma part of the mechanism for clinical application experimental basis. Methods: The rats were randomly divided into normal control group (A), asthma model group (B), dexamethasone treatment group (C), the God vanilla high-dose treatment group (D), God vanilla low-dose treatment group (E) . Joint sensitization and repeated atomization method for preparing asthma model. Number of counting the rats in bronchoalveolar lavage fluid (BALF) in eosinophils (EOS); enzyme-linked immunosorbent assay (ELISA) to detect serum eosinophil chemotactic factor-2 ,3 ( Eotaxin-2, Eotaxin-3), the level of soluble P-selectin (sP-selection), interleukin-17 (IL-17); lung tissue by reverse transcription polymerase chain reaction (RT-PCR) detection signal transduction guide and activator of transcription factor 3 (STAT-3), retinoic acid nuclear orphan receptor γt (RORγt), IL-17mRNA the expression levels. Results: 1) compared with the control group, asthma model group, the number of EOS in BALF increased significantly (P lt; 0.05), serum of Eotaxin-2, Eotaxin-3, SP-Selection and IL-17 level was significantly higher (P lt; 0.05), the lung tissue STAT-3, RORγt and IL-17 mRNA expression was significantly increased (P lt; 0.05); 2) compared with the asthma model group, the treatment groups EOS in BALF significantly reduced the number serum eotaxin-2 and IL-17 levels were significantly decreased (P lt; 0.05), with God vanilla low-dose treatment group compared to the number of EOS in the high dose group in BALF significantly reduced serum eotaxin-2 and IL-17 levels were significantly decreased (P lt; 0.05); 3) in serum of Eotaxin-2 level the BALF the EOS number of in closely related (r = 0.829, P lt; 0.05), serum Eotaxin 3 and SP-selection level in the number of EOS in BALF were positively correlated (r = 0.635,0.348, P lt; 0.05); 4) the serum level of IL-17 serum Eotaxin-2, Eotaxin-3 was a positive correlation between (r 0.613,0.491,0.350, P lt; 0.05); 5) compared with the asthma model group, the treatment groups in rat lung tissue STAT-3, RORγt, IL-17 and SP-selection level mRNA expression levels were significantly decreased (P lt; 0.05), with God vanilla low-dose treatment group compared to the high-dose treatment lung tissue STAT-3, RORγt, IL-17 mRNA expression level was significantly decreased (P lt; 0.05); 6) the rats lung tissue STAT-3 mRNA expression levels of IL-17 mRNA expression level was positively correlated (r = 0.976, P lt; 0.05) RORγt mRNA expression levels of IL-17 mRNA expression levels were closely positively correlated (r = 0.983, P lt; 0.05), STAT-3 mRNA of the expression level of RORγt mRNA expression level was positively correlated (r = 0.939, P lt; 0.05) Conclusion: of Eotaxin and SP-selection can promote EOS inflammatory infiltrate in the airway; IL-17 involved in the pathogenesis of asthma and in collaboration with other role of proinflammatory cytokines; STAT-3 and RORyt can promote asthmatic rats IL-17 expression; dimensional god of medicine herbs can reduce asthma serum of Eotaxin-2 level, reducing the EOS infiltration of the airway; God vanilla can reduce asthma serum IL-17 level and inhibit asthma Rat lung tissue STAT-3 and RORγt expression, thereby inhibiting the expression of IL-17. This may be one of the mechanisms of God vanilla reduce asthma airway inflammation in rats.
