Abstract

This paper is divided into two parts, the first part of the preparation of synthetic peptide antibody investigated the relationship between the different organs of mung bean seedlings cyanide-resistant respiration activity of alternative oxidase expression; second part of the different periods of vernalization cyanide-resistant respiration under winter wheat germ pathway and alternative oxidase expression variation. First portion in accordance with the alternative oxidase (Alternative Oxidase, AOX) S-helical amino acid sequence is located in the hydrophilic region of the outside of the inner mitochondrial membrane, with a solid-phase synthesis of a polypeptide consisting of 12 amino acid residues of this polypeptide with α- Bovine chymotrypsin is connected to the original A, used as a hapten, the antibody prepared by immunizing rabbits synthetic 12 peptide. This antibody with the total protein from mung bean seedlings different organs mitochondria and Western blotting, respectively, can be observed in mitochondria of mung bean seedlings having two clear of AOX hybridization band, they are the molecular weight of 35kD and 38kD respectively. The same time, take the cotyledons of mung bean seedlings, leaf and hypocotyl were measured and calculated their respiratory parameters, you can learn the true leaf organs and tissues has the largest total respiration (V t ) and the alternative pathway the actual operation of the capacity (V alt ) and the alternative pathway amount (ρ V alt ); The Second is the cotyledons V t and ρ V alt ; hypocotyl V t and ρ V alt , the lowest, but the V alt is higher than the cotyledons. On respiratory parameters of the different organs of mung bean seedlings results AOX expression was measured by Western analysis basically the same: especially true leaf of V t ρ V alt highest, it also has a 35kD 38kD The AOX hybrid polypeptide; followed cotyledons V t and ρ V alt , cotyledons, saw a molecular weight of 38kD AOX polypeptide; hypocotyl the V t and ρ V alt , are lowest, Western blotting showed that only a molecular weight of 38kD polypeptide expression levels less. The 35kD The AOX might Leaf of ρ V alt , made a major contribution. The second part of winter wheat materials, different periods of low temperature treatment under the conditions of the non-stress vernalization Abstract winter wheat germ cyanide-resistant respiration, run and alternative oxidase expression change. The results show that: winter wheat germ in the early days of vernalization, along with the rise in the total respiration rate, and the rapid increase in the cyanide-resistant respiratory capacity (Valt) and the actual running activity (p Valt), but showed a downward trend in the vernalization late both is very consistent with the results of Western blotting; throughout the vernalization period, despite increased activity of the cyanide-resistant respiration run or at a lower level, but relative to the for cytochrome main road, the main road of cytochrome continues to occupy the advantage. Accordingly, we believe that vernalization breathing against cyanide induced only performance at an early stage, and to adapt to the cold of winter plant wintering early for.
