Experimental Studies on Spermatogenic Cells Apoptosis in Rats with Varicocele and Soluble Granule of Sheng Jing Influencing the Apoptosis

Abstract


Objective: In this paper , by observing the experimental varicocele rat spermatogenic cell apoptosis index ( AI ) , testicular weight , varicose vein diameter , testicular histological changes , apoptosis of spermatogenic cells of rat testicular varicocele the condition and spermatogenic granules varicocele rat testicular spermatogenic function of the protective effect . Methods: Adult male Wistar rats ( 310 ± 10g ) , 60 only on the the Ishizu , Kazuhiko improved legal system into the left varicocele rats were randomly divided into control group ( n = 20 ) ( sham-operated group SOG ) , model group ( n = 20 ) ( varicocele VG ) , treatment group ( n = 20 ) ( modeling traditional Chinese medicine VMG ) . Treatment group a week after surgery to spermatogenic granules ( 1.2g/300g ) once a day orally ; model group, a week after surgery , and to the dose of water once a day orally . Each group fed normal diet after three months , the surgical testicular weighing ; the HE staining histopathology observed ; situ terminal deoxynucleotidyl transferase labeling ( TUNEL ) detection of testicular germ cell apoptosis . Results : ( 1 ) VG rat left spermatic vein diameter than the SOG significantly the varicose ( P lt ; 0.001 ) , VMG rat left spermatic vein diameter and the VG was no significant difference ( P gt ; 0.05 ) ; ( 2 ) VG left testicle weight was significantly lower than the SOG and VMG ( P lt ; 0.001 ) , VMG and SOG was no significant difference ( P gt ; 0.05 ) ; ( 3 ) VG the left testicle AI significantly higher than SOG ( P lt ; 0.001 ) , VMG ( P lt ; 0.01 ) , VMG and SOG were significantly different ( P lt ; 0.01 ) ; ( 4 ) VMG histopathological changes significantly lighter in VG , VMG song seminiferous tubules basement membrane thickening , stenosis and late in the process of spermatogenesis interrupt and other pathological changes is not obvious. Conclusion: experimental varicocele testis of rats pathological damage ; increase in apoptosis of spermatogenic cells , which may be one of the mechanisms affecting fertility ; the spermatogenic granules can reduce the varicocele rat spermatogenic cells withered death, has a protective effect on spermatogenesis , thereby increasing the reproductive capacity of the testis .
