Objective: To observe octreotide (octreotide, Oct) transcatheter arterial chemotherapy via the hepatic artery infusion chemotherapy (TAC) combined with portal vein infusion chemotherapy (portal vein chemotherapy, PVC) treatment effect on implanted liver cancer in rats; octreotide with fluorouracil (5-fluorouracil ,5-Fu) local infusion treatment efficacy of implanted liver cancer in rats. Provide a theoretical basis for clinical. Methods: 30 male Sprague-Dawley rats were randomly divided into three groups, each of 10 A group for the experimental group, B group, negative control group, positive control group, C group. Walker-256 cell lines grown in the rat liver group A, B, C, the TAC PVC program perfusion Oct, 5-Fu and normal saline (the normal saline NS), 7 days after the rats were sacrificed, observe and compare groups The change in tumor volume, apoptosis index. Replacement of 30 male Sprague-Dawley rats, grouped Ibid, comparison of survival time after reperfusion. : A, group B dual 7 days after reperfusion average inhibition rate of 62.12%, 59.44%, respectively (P gt; 0.05), A, group B and group C tumor volume was significantly different (P lt; 0.05). Immunohistochemistry by TUNEL: A, B and C, compare the difference was significant significance (P lt; 0.01); A and B compare the difference was significant (P lt; 0.01). Immunohistochemistry of Bax: A and C compare the difference significant significance (P lt; 0.01); difference between B and C compare non-significant with significance (P gt; .05); A and B lt; WP = 7 gt; compare the difference there is significant significance (P lt; 0.01). Immunohistochemical Bcl-2: A, B and C compare the difference was significant (P lt; 0.01); A and B compare the difference was significant (P lt; 0.01). Compared in adjacent normal liver tissue: A and B group TUNEL, Bax, expression of Bcl-2 staining and expression intensity were significant difference (P lt; 0.01). Electron microscopy: A group of visible obvious change in nuclei of tumor cells and mitochondrial apoptosis, apoptotic changes than group B Group A slight, while Group C of apoptotic changes rare. After the longest survival time of 39 days, the octreotide group; postoperative shortest survival time of 22 days for the saline control group. Compare the survival time of the group A and group C, the difference was significant (P lt; 0.05); between group A and group B, between B and C groups survival time was no significant difference significance (P gt; .05). Conclusion: Octreotide by local perfusion can inhibit the growth of implanted liver cancer in rats, and promote the occurrence of apoptosis in cancer cells; compared with 5-Fu antitumor effect of octreotide is more obvious, and the more minor damage to the normal liver tissue. Octreotide after local perfusion treatment to extend the survival time of rats with implanted liver cancer; compared with 5 - fluorouracil, octreotide by local perfusion effect no significant difference in the survival time of rats with implanted liver cancer.
