Effects of Botulinum Toxin A on Rat Model of Benign Prostate Hypertrophy

Abstract


Objective: To investigate the type A botulinum toxin ( BTX - A ) on rat prostate hyperplasia model . Methods: 64 male Wistar rats were randomly divided into model group ( 52 ) , sham operation group ( 12 ) . Model set of rows 1 week after castration , to subcutaneous injection of testosterone propionate 4mg/d.kg modeling , for 1 week . Sham-operated group over the same period to the subcutaneous injection of the same amount of olive oil , for 1 week . Random to 4 model mice were sacrificed , remove the prostate , light microscope confirmed successful model . The remaining 48 model rats were randomly divided into high , low dose group, negative control group , each group of 12 , respectively, to the group of prostate injection 15u/0.1ml in BTX - A , 10u/0.1ml BTX - A, 5u/0.1ml BTX-A, 0.1ml saline . Continue to subcutaneous injection of testosterone propionate 4mg/d.kg modeling for four consecutive weeks ; sham operation group continues to subcutaneous injection of the same amount of olive oil , for four consecutive weeks . Each group of mice were injected two weeks after 4 weeks in 2 batches ( six each ) were drawn in the prostate , bladder , heart , kidney , liver , light microscope to observe the morphological changes , calculated for each experimental group prostate weight , the prostate index ( PI ) . Gland area of the semi-quantitative detection of the pathological image analysis system , interstitial area . Immunohistochemical detection of bax , fas , bcl - 2 protein expression . All data were analyzed using SPSS package . Results: Compared with the negative control group , administered two weeks , the low-dose group prostate weight , PI , glandular area , bcl-2 expression decreased ( P lt ; 0.05 ) ; Bax , Fas expression was increased ( P lt interstitial area ; 0.05 ) ; relative increase ( P lt ; 0.05 ) . Compared with the negative control group , administered four weeks , the middle dose group prostate weight , PI , glandular area , bcl-2 expression decreased ( P lt ; 0.05 ) , Bax , Fas expression increased ( P lt ; 0.05 ) interstitial area relative increase ( P lt ; 0.05 ) . Gland area of the low-dose group decreased ( P lt ; 0.05 ) , interstitial area compared to the low-dose group and negative control group , the difference was not statistically significant ( P GT ; .05 ) . 2,4 weeks of the indicators of the sham-operated group , the difference was not statistically significant ( P gt ; 0.05 ) Conclusion : BTX - A lead to the proliferation of rat prostate gland volume reduction , is expected to become a new method for the clinical treatment of benign prostatic hyperplasia .
