Abstract


Zanthoxylum (Zanthoxylum L.) is the most widely distributed Rutaceae (Rutaceae) a genus of about 250 species worldwide, widely distributed in Asia, Africa, Oceania, North America, tropical and subtropical regions, temperate distribution less. China has 39 species and 14 varieties are produced in North and South, mainly distributed in the provinces and regions south of the Yangtze River and its; 18 in Sichuan and Chongqing, 8 variants, one variant. Most species of this genus, roots, stems, leaves, fruit can be used as medicine, analgesic, anesthetic, antibacterial, anthelmintic and anti-cancer effects, the fruit is often used as a flavoring agent and preservative, has a higher economic value ; important tree species for reforestation of mountain ecosystems while current returning farmland to forest project as an important ecological and economic species. From both morphological and molecular biology level, combined with a wide variety of statistical analysis software, are plant parts interspecific and intraspecific genetic relationship for the first time in Sichuan and Chongqing pepper. For species in order to system research, of germplasm resources development, utilization and protection to provide a theoretical basis. The results are as follows: 1 pepper morphological study of the use of SPSS data analysis software is a kinship, the systematics relationship of Sichuan and Chongqing pepper genus taxa Q-type and R-type cluster analysis and principal component analysis. (1) 44 traits indicators Zanthoxylum farthest neighbor-joining method pepper are 16 kinds, Q-6 variant and a variant clustering. Clustering results show that the the: most types existing Zanthoxylum classification system consistent heterophylla the pepper and SPINIFOLIUM Zanthoxylum attribution greater differentiation; heterophylla pepper is a pepper subgenus traditional classification system, this paper poly class results showed that the the the different leaf pepper and Cliff pepper subgenus close relationship with the pepper Asia is a distant relationship between the different groups had. (2) R cluster of traits: test selected 44 traits are independent of each other traits, the number of tepals, perianth differentiated into calyx and petals, female flower style whether the separation of the three great correlation who perianth differentiated into sepals and petals types of female flowers style separation and the tepals number the same. The most classification group traits more complicated, lack intuitive and regularity, reflects this characteristic pepper variation of individual traits. (3) The principal component analysis showed that: some traits there is a large correlation. The principal component analysis to take cumulative contribution rate of greater than 85% of the nine main ingredients, the cumulative contribution rate of 85.519%. I principal component contribution rate of 25.748%, reflects the leaf morphological characteristics. Principal component Ⅱ contribution rate of 14.850 percent, reflecting the blade shape and composition of the main components. Principal component Ⅲ contribution rate of 10.725%, to reflect the characteristics of the plant branches and leaves appendages. The principal component Ⅳ contribution rate of 8.844% to reflect the characteristics of the the petiole, Yu handle glands. These main ingredients difference basically reflects the property of their respective kinds of typical characteristics of the pepper. 2 pepper genus phylogenetic relationship of RAPD analysis (1) from 62 random primers material screened out of the 15 can be amplified by a clear strip with the primers, 15 primers in the 11 copies of the test material was amplified out of 244 with an average of primers amplified with 16.3. Of polymorphic bands of 243, 99.6% of the total amplified bands number. The size of the amplification product in the 396-3000 bp range, mainly between 1000-1600bp. (2) According to the PCR results, with the NTSYS-pc2.02a 11 copies of the test material between the genetic similarity coefficient. The similarity between the various types of coefficient of variation range of 0.506-0.922, the average was 0.617. UPGMA method based on similarity coefficient, clustering, clustering branching tree. The results show that, the pepper together the case of most of the species and its variants or variant kinship than intraspecific interspecific basically consistent with the results of the traditional classification. The larger difference is the Zanthoxylum SPINIFOLIUM pepper subgenus cliff pepper subgenus relationship closer, the number of classification results with morphological traits consistent. 3 Zanthoxylum Zanthoxylum calyx and petals of the species included Cliff pepper subgenus classification system for analysis of the traditional classification, no sepals and petals divided species classified pepper subgenus. In this study, the number of analysis and RAPD analysis results showed that belongs to the Zanthoxylum subgenus SPINIFOLIUM Zanthoxylum cliff pepper subgenus relationship compared pepper subgenus nearly leaves pepper classification of SPINIFOLIUM larger differences other species divided traditional classification results are basically the same. Comprehensive pepper is a variety of morphological characteristics between the variants and RAPD analysis, preliminary taxa included in subgenus of pepper under the adjustment, the first time the case is divided into five groups: 1) pepper Group, pepper, The bamboo leaves pepper and its variants, or variants, burrs, pepper, pepper Sichuan and Shaanxi, slightly pubescent pepper; 2) group of Shell pepper, Shell pepper its variants; 3) large flower pepper group, different leaf pepper, the thorns shell pepper, large flower pepper ; 4) the Leibo pepper group, Leibo pepper alone group; 5) flowers pepper, pepper, Guizhou, narrow leaf pepper, the Bashan pepper, water chestnut leaves pepper, the pepper ilicifolius and flowers pepper. The study shows that the division pepper genus subgenus traditional classification groups based primarily on the differentiation of sepals and petals and female styles whether separation In this study, the number of classification, a combination of the pepper is a large number of morphological traits to a greater extent to overcome the limitations of traditional morphological classification, while the use of RAPD markers to verify the results more reliable.
