The Role of PMNs, Mononuclear Macrophage and GM-CSF in Vaginal Candidiasis

Abstract


Objective: The purpose of the thesis is to study the innate immunity in vaginal candidiasis by murine model of vaginal candidiasis, through detecting the content variation about polymorphonuclear neutrophils (PMNs), mononuclear macrophage and granulocyte-marcophage colony stimulating factor (GM-CSF) and cfu in blood and vaginal fluids during the whole infection. Also some models are treated by recombinant human granulocyte-macrophage colony stimulating factor( rhGM-CSF) .Method: Seventy-two femal Kunming mice, 8-10 weeks of age ,are used throughout these studies. All animals are randomly divided intofive groups. Group A is infected only once and group B is infected two times. Separate groups of mice are detected the content variation about PMNs, mononuclear macrophage and GM-CSF and cfu in blood and vaginal fluids on days 2,7and 14 postinfection. Group C and group D of mice are separately treated by sporanox through intragastic administration and by rhGM-CSF through intraperitoneal injection on days 2 postinfection, which are repeated for 3 days. The last two groups of mice are detected the content variation about polymorphonuclear neutrophils and mononuclear macrophage in blood and vaginal fluids , and cfu in vaginal fluids on days 3 after treatment. Group E is normal. The results are processed by SPSS 11.0 statistic software.Results: The content variation about PMNs, mononuclear macrophage, GM-CSF and cfu in vaginal fluids and blood are highest on days 7 in both the first and the second group. According to the same days postinfection, the content variation of the second group about PMNs, mononuclear macrophage, GM-CSF and cfu in vaginal fluids and blood,which are higher than of the first group, also the content variation in vaginal fluids is higher than in the blood.The mice who are treated by sporanox through intragastic administration, whose vaginal fluids are not detected PMNs and mononuclear macrophage, content variation of PMNs and mononuclear macrophage in blood are lower than those of the mice who are not treated by any medicine, and the eight mice are all cured. The mice who are treated by rhGM-CSF through intraperitoneal injection, whose content variation of PMNs and mononuclear macrophage are higher than those of the mice who are not treated by any medicine in both blood and vaginal fluids,and there are six mice are cured in this group. All the index in vaginal fluids of the fifth group is not detected, while content variation of all the index in the blood is lower than the other groups,except the third group. Conclusion1. The content variation of PMNs and mononuclear macrophage is related with symptomatic infection in this model, especially the content variation in vaginal fluids.2. PMNs and mononuclear macrophage and GM-CSF may stimulate that mice counteract the infection of vaginal Candida albicans3. Recombinant human granulocyte-macrophage colony stimulating factor can promote content variation of polymorphonuclear neutrophils and mononuclear macrophage in both blood and vaginal fluids, and the local role is more important than systemic role.
