Abstract


Background: Ovarian carcinoma is one of the tumors which are most difficult to cure. Although much improve have taken place in surgery,chemio-and radiation therapy for recent years, the five-survival rate of adcanced ovarian carcimoma remains low .It is therefore obvious that other therapies must be developed to prolong survival of patients with advanced disease. Recently ,Along with the development of cell and gene engineering techniques, gene therapy have progressed and become perspective in antitumor therapy. Interleukin-24(IL-24) is a relatively new cytokine and is regarded as inhibiting the proliferation of human cancer cells, but have no effect on normal cells, it has become the hot spot. there was nearly no research about gene therapy of IL-24 in ovarian carcinoma. We also have constracted the reconbinant eukaryotic expression vectors pcDNA3.1(—) IL-24 .On this basement,we designed this experiment.Objective: To analyze the role of human IL-24 gene on the proliferation of human ovarian carcinoma cell line(SKOV3).Methods: The IL-24 expression vector pcDNA3.1(—) -IL-24 was transfected into SKOV3 by electroporation .The transfected cells were screened with G418,and pcDNA3.1(—) -IL-24 mRNA were tested byRT-PCR. At the same time, pcDNA3.1(—) were transfectd into SKOV3 as empty-vector control,wild-grow SKOV3 as negative control, the proliferation of SKOV3 was tested by MTT assay.Result:1. IL-24 transfected group ( SKOV3 transfected with pcDNA3.1(-)-IL-24) and empty- vector group (SKOV3 ransfected with pcDNA3.1(-)) SKOV3 cells continued growing in G418 selection culture media and the G418 resistance were remained 2 weeks at least , whereas negative control group SKOV3 cells (nontransfected SKOV3 cells)all died in this mesia.2. IL-24 mRNA and GADPH were tested by RT-PCR. IL-24 transfected group showes positive results with specific clip for IL-24 mRNA,whereas empty- vector group and negative control group . The relative expression values of mRNA showed zero in empty- vector group and negative control group ,and 0.18±0.03 in IL-24 transfected group.3. The OD values showed significant difference among three groups of cells by MTT assay(p<0.001).The growth rate of IL-24-transfected cells was inhibited,while the cell growth curve of empty-vector group was similar to that of nagative control group.Conclusion:1. IL-24 can inhibit the growth of SKOV3 cell in vitro.2. There may be a potential value of IL-24 gene therapy for ovarian carcinoma.
