Abstract


Objective:To investigate the expression of LIFmRNA and LIFRmRNA in the ovaries of the patients with polycystic ovarian syndrome(PCOS) and the relationship between them and the apoptosis of ovarian cells female hormones, to explain further the mechanism the failure of follicular development for LIFmRNA and LIFRmRNA in PCOS.Methods: Collecting a bit of ovary of 20 patients with PCOS and 20 control patients,detecting the expression of LIFmRNA and LIFRmRNA with in situ hybridization histochemistry(ISHH) and examining the apoptosis of cells by TdT mediated dUTP nick end labeling (TUNEL).Result:1.The expression of LIFmRNA and LIFRmRNA were found in both granulosa cells and intersititial cells,the staining for the former was stronger than the latter (P=0.011 in the study group, P=0.000 in the control group) . LIFmRNA and LIFRmRNA were kept consistent in the control group(P=0.930 in granulosa cells, P=0.904 in intersititial cells).The intensity of LIFmRNA in granulosa cells of the study group was significantly weaker than the control group (P=0.019) while the expression of LIFRmRNA had no variety (P=0.857) .In two groups the intensity of staining for LIFmRNA and LIFRmRNA in granulosa cells increased as follicles became larger, whereas the expression of LIFmRNA in all size of follicles of the patients with PCOS was observed lower than normal ovaries(P=0.000).2. In the study group apoptosis index (AI) of granulosa cells was obviously higher than the control group(P=0.000) and oocytes did not appear apoptosis except atretie follicle. In two groups AI of granulosa cells decreased as follicles became larger, whereas in the study group AI of granulosa cells of all size of follicles was observed higher than normal ovaries(P=0.000) and the expression of LIFmRNA correlated negatively with AI in granulosa cells(P=0.000, r=-0.796).3. The mean level of serum LH, LH/FSH and T of PCOS patients was higher than the control patients(P=0.000,0.000,0.008) and the expression of LIFmRNA in granulosa cells correlated negatively with E2(P=0.000,r=-0.771) and T (P=0.002,r=-0.656).Conclusion:1. LIFmRNA and LIFRmRNA exist in ovarian granulosa cells and intersititial cells, dyregulation of LIFmRNA and disruption of LIFRmRNA may be one of the pathogenesis of PCOS.2. Dyregulation of LIFmRNA probably increase apoptosis of granulosa cells through a certain path so that follicles stop growth and the patients with PCOS can not ovulate.3. Female hormones and LIFmRNA perhaps adjust each other and their abnomal regulation may cause PCOS.
