Abstract


Background: Cytieochrome P450 was named in 1961,because the pigment (P) has a450-nm spectral peak when reduced and bound to carbon monoxide. According to the develop-ment of molecular biology technology, cloning studies have revealed dozens of different CYPenzymes, which comprise a superfamily of hemethiolate proteins. In humans, CYP enzymes playa central role in PhaseⅠdrug reactions, which involved in many drug interactions, and contributeto interindividual variability in drug metabolism. The CYP enzymes are divided on the basis ofamino acid identity into families (enzymes that share≤40% identity) and subfamilies (sharing≥55% identity). In the nomenclature system, the symbol CYP representing cytochrome P450 isfollowed by an Arabic numeral denoting the family. a letter designating the subfamily, and anArabic numeral representing the individual gene within the subfamily, for example CYP1A2. Inhuman beings, 57 CYP genes and 33 nonfunctional pseudogenes have been arranged into 18 fami-lies and 42 subfamils.CYP1A2, in contrast, is constitutively expressed in the liver where it constitutes about 13%of total liver CYP content,.which preferentially metabolizes arylamines and N-hetero-cyclics, andis involved in metabolism of a number of drugs such as caffeine, clomipramine, clozapine andlocal anaesthesia drug such as lidocaine and ropivacaineo CYP1A2 exist genetic polymprphismand have different in race,sex and age. The study of genetic polymorphism of CYP1A2 is verymore in whiter of Europe,Japanese,and Han population of China in Asia. China have 56 folks.Dai population mainly settle in Banna and Dehong territory of YunNan province. The aim of this shudy is mainly confirmation the enetic polymprphism of CYP1A2 in Dai and Han population.Aim: We use polymerase chain reaction-restriction fragment length polymorphism toanalyze the single-nucleotide polymorphism (SNP)G2964>A and frequency of CYP1A2 of Daipopulation and Han population in Dehong territory of YunNan province.Methods: 229 unrelated Chinese healthy subjects who live in Dehong territory YunnanProvince,including 112 Han (male 60,female 52),and 117 Dai (male 42,female 75),were enrolledin this study. Polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP)was employed in genotyping.Result: The allelic frequencies of CYP1A2 2964 location in Dai and ban populationswere 0.34 (95% CI:0.3-0.4) and 0.24 (95% CI:0.2-0.3) respectively. The incidences of A2964 inDai was higher than that in Han (p<0.05).The distribution of the genotypes in both the populati-ons was in Hard-weinberg equilibrium with respect to G2964A of CYP1A2. The incidence of2964G/A genotypes were significance between the two populations(X2=7.411 df=2 p<0.05).Conclusion:1. There are ethnic differences in G2964A genetic polymorphism of CYP1A2 in Han populationand Dai population in Dehong territory of YunNan province.2. There are statistical difference in genetic frequency of CYP1A2 in Han population and Daipopulation in Dehong territory of YunNan province.3. There are statistical difference in genotype (G/G, G/A, A/A) of CYP1A2-2964 site inHan population and Dai population in Dehong territory of YunNan province.4. The drug metabolismed by CYP1A2 enzyme maybe have difference in Han population andDai population in Dehong territory of YunNan province.
