Abstract


Mungbean(Vigna radiata (L.) Wilczek)belongs to Ceratotropis and it is popularly planted in south Asia. Bruchid is a main pest of mungbean and can bring great harm to mungbean production and storage. So, resistance variety is an important way to prevent bruchid. In present study, the major objectives were to study the inheritance of resistance to the cultivar mungbean, V2709 and to tag the resistance gene by molecular markers, in order to provide useful information for resistance breeding and fine mapping of resistant gene. The main results were as follows:1. By using the bruchid resistant cultivar V2709, the susceptible cultivar VC1973A, and their F2, F3 to study the inheritance of bruchid resistance in V2709. The results were as follows: F1 individuals were all resistant to bruchis, the segregation in F2 and BC1F1 population were 3R:1S and 1R:1S respectively. F3 population segregated in a 1:2:1 ratio. It indicated that the bruchid resistance of V2709 is mainly controlled by one dominant major gene.2. Five mungbean accessions with resistance to bruchid, V2802, V2709, VC6089A-6, ACC41, and VC1973A/V2709-2, were crossed with each other to test the allelism of the resistance genes. Segregation in the F2 populations of bruchid resistance from (VC1973A/V2709-2)×ACC41, VC6089A-6×V2709 and VC6089A-6×V2802 were suggested that the resistance genes between VC1973A/V2709-2 and ACC41; between VC6089A-6 and V2709; between VC6089A-6 and V2802 were non-allelic while the segregation in F2 populations of the crosses (VC1973A/V2709-2)×V2802, (VC 1973A/V2709-2)×V2709 indicated that the resistant gene in them are allelic or closely linked.3. At the same time, 63 RAPD markers, 100 SSR and 28 STS primers were used for BSA analysis to find molecular marker that link with the resistant gene. As the results, OPC-06 and STSbr2 were identified to be related with the bruchid resistant gene. After verified in F2 individuals, the genetic distances between these two markers and the target gene were 11.0cM and 5.8cM respectively.
