Dermatophilus congolensis is a member of Actinomycetales,Dermatophilaceae, Dermatophilus. Caused the diseases of an acute or chronic on a variety of animal and human by the skin contanct. With superficial exudative pustule rash or nodules as the main clinical features. The infection may cause a decline in productivity and resistance of disease, infection on Sheep can lead to reduce the quality of wool and leather, OIE lists the disease in B class.Because the characteristics of the growth of Dermatophilus congolensis in spores and hyphae. Dermatophilus congolensis isolated from skin lesions is often difficult to obtain pure culture bacteria. Therefore the establishment of an efficient and effective separation of the Dermatophilus congolensis.To isolated and identificate of Dermatophilus Congolensis of Argali sheep.In this study, purified and identified sheep strains of Dermatophilus congolensis as material,the different pretreatment methods and the culture medium was used for the isolation of Dermatophilus congolensis. established a effect method was for laboratory experiment.Results show that:1. The skin material was dried at 28℃and smashed, added with sterile water for 15 days, standing.Added with 6% yeast extract was stirred at 60℃for 30 min and scribbled on plates at 37℃for 48 h, to evaluate the characteristics of the Dermatophilus Congolensis,37℃water bathed 1h, or placed in candle 15min isolated no bacteria; 0.05% SDS and the 0.05% SDS 6% protein peptone is addicted to the activation of the bacteria;The bacteria on Gauze’medium No.1 poor growth; LB agar is inferior to sheep blood agar.2. The bacteria collected from Argali sheep were related to the Dermatophilus Congolensis. the bacterial morphology, cultivating characteristics, indirect immunohistochemistry were performed and devised a pair of specific primers according to the 16S rRNA sequence of Dermatophilus Congolensis GenBank published for PCR.3. The result of DNA random amplified polymorphic DNA (RAPD) analysis showed that two different hosts originate Dermatophilus Congolensis have different genome.
