Abstract


Preface zinc ion is one of the important trace elements in the body, is an important part of a variety of enzymes and proteins. In the central nervous system, the zinc ion content is extremely rich. A large number of studies have shown that free zinc ions as a neuromodulator, coexist in a zinc-containing neurons axon terminals synaptic vesicles with glutamate and other neurotransmitters in the synaptic activity neurotransmitter released into the synaptic cleft, to participate in the regulation of neurotransmitter release and NMDA, AMPA, GABA and glycine receptor function. As part of the central nervous system, retinal, also contain a certain concentration of zinc ions, zinc ions may have an important role in the regulation of the related functions of the retina. Recent studies show that, closely related to the zinc ion transmembrane transporter protein family zinc transporter (zinc transporter, ZNT) play a key role in the maintenance of intracellular zinc homeostasis process. Zinc transporter family of seven members, namely ZNT1-7, already have been cloned, and there is a certain understanding of its function. It has been reported ZNT3 expression in the retina retina there strong zinc metabolism. However, about free zinc ions in mouse retina and the positioning of the zinc transporter family members addition to ZNT3 outside distribution and no detailed report. Therefore, this study adopts the specific detection of free zinc ions of metal autoradiography (autometallograpby AMG), immunohistochemistry and Western Blot technology of free zinc ions in mouse retina and six zinc transporter (ZNT1 ZNT3 ZNT4 ZNT5, ZNT6, ZNT7) positioning distribution and expression were studied in detail, and lay a morphological basis to further investigate the zinc ion and zinc transporter in the physiological function of the retina. Materials and Methods 40 adult male CD-1 mice, weighing between 40-50g, were housed separately regular diet and tap water supply. The application of AMG technology, immunohistochemistry and Western Blot Detection of mouse retina free zinc ions and zinc transporter ZNT1, ZNT3 ZNT4 ZNT5 ZNT6, ZNT7 positioning distribution and expression. The results the Free Zinc ions and zinc transporter ZNT1 ZNT3, ZNT4 ZNT5 ZNT6 ZNT7 widely present in the mouse retina layers. Free zinc ions are mainly distributed in the mouse retinal pigment epithelial cell layer, photoreceptor inner segment, outer nuclear layer, outer plexiform layer, inner nuclear layer, inner plexiform layer and the ganglion cell layer. Zinc transporter in retinal layers distribution: ZNT1 and ZNT6 distribution similar strong positive reaction to exist within the protrusions of the outer plexiform layer and inner plexiform layer, nerve fiber layer, ganglion cell layer and pigment epithelial cell layer moderate positive reaction, pigment epithelial cell layer, the photoreceptor inner section and the outer section ZNT6 significantly stronger ZNT1, both in the inner nuclear layer and outer nuclear layer was no significant response. ZNT3 strong diffuse positive reaction in the retina, in the outer segment of the photoreceptors, pigment epithelial cell layer, outer plexiform layer and the inner plexiform layer. ZNT4 and ZNT7 exists a strong positive reaction to moderate positive kernel layer visible part of the positive reaction of the cells within the ganglion cell layer and pigment epithelial cell layer, outer plexiform layer and the inner plexiform layer protrusions, but rare in the outer nuclear layer, the second in the positive reaction of the inner segment and outer segment of the photoreceptors are weak. ZNT5 the strong positive reaction mainly exists in the photoreceptors in the outer section, ganglion cell layer, inner nuclear layer of photoreceptors inner segments and pigment epithelial cell layer also have significant positive reaction, outer plexiform layer and within the network layer of the projections within the positive reaction diffusion distribution of the outer nuclear layer of positive response is not obvious. Conclusion mouse retina contains a wealth of free zinc ions and zinc transporter ZNT1 ZNT3 ZNT4 ZNT5, ZNT6 ZNT7 prompted as the neuromodulator zinc ion conduction and the formation process of retinal neurons in the visual information may play important role, six kinds of zinc transporter may be detected in mouse retina, play an important role in the metabolic process in zinc.
