Abstract


Petroleum hydrocarbons are widespread industrial pollutants that are released into the environment by transporting and storing of crude oil, refining of petroleum, accidental leaking and petroleum-containing wastewater irrigation, etc. However, microbial diversity impacts of petroleum pollution on soil by ARDRA have been scarcely studied.Conventional culture method and modern molecular techniques 16s rDNA polymerase chain reaction (PCR)- Amplified-rDNA Restriction Analysis(ARDRA) were combined to evaluate the effects of long-term petroleum-containing wastewater irrigation on bacterial community structures and diversities in soils in Shenfu irrigation area, the largest area irrigated by oil-containing wastewater for more than 50 yr in northeastern China. The results showed that soil total petroleum hydrocarbon (TPH) and polycyclic aromatic hydrocarbons(PAHs) concentration declined along the gradient of the irrigation channel from up- to downstream. Construction of 16s rDNA genomic library had population diversity. Proteobacteria and Actinobacteria were dominant in Shenfu irrigation area. Also contained many kinds of survived bacteria but uncultured.Three pyrene-degrading strains and one fluoranthene-degrading strain were isolated by selective enrichment culture and screening model of an overlayer from high concentration diesel oil-stressed soil in laboratory. The effect of co metabolic in increasing the degradation effeciency of PAHs had been studied preliminarily.
