Experimental Study on the Early Intervention of Hormonally Ischemic Necrosis of Femoral Head in Rabbits with Combined Use of Atorvastatin and Clopidogrel

Abstract


Objective To study the effects of blood viscosity and blood fat of hormonally osteonecrosis of femoral head in rabbits with combined use of Atorvastatin and Clopidogrel.To investigate the feasibility and mechanism of precaution and treatment of steroid-induced avascular necrosis of the femoral head with these two drugs.Method Twenty-four adult New Zealand white rabbits were divided randomly into three groups:A,model group(n=8);B,treatment group(n=8);C,control group(n=8).Model group and treatment group were injected twice with 10ml/kg/time equine serum into ear-vein every 4 weeks.And then they were injected intraperitoneal with 10mg/kg.d of dexamethasone sodium phosphate for 3 days.After that,treatment group was intragastric administration with 4rag/kg.d of Clopidogrel and 4rag/kg.d of Atorvastatin.From the fifth week,the control group was injected intraperitoneal with 4mg/kg.d of physiologic saline for 3days.Before and after the experiment at 5,6,8,10 weeks,serum cholesterol,triglyeride and plasma viscosity were tested. We observed the necrosis of samples through X-ray and pathological section.Results The result display that serum cholesterol,triglyeride and plasma viscosity of model group and treatment group were higher than control group;and the model group was higher than the control group.There is significant difference between every two groups(P＜0.05). It demonstrated that early intervention with Atorvastatin and Clopidogrel was valid.We can see the incidence rate of SANFH of treatment group is obviously lower than model group through X-ray and pathological section,and there is no ONFH in control group.Conclusion It can induce osteonecrosis of the femoral head of the adult rabbits to use large dose hormone.It decreased blood viscosity and fat metabolic disorder to combined use of Atorvastatin and Clopidogrel,and maybe contributed to prevent SANFH.
