The main purpose of this topic is to explore the use of high-speed countercurrent chromatography separation and purification, HPLC analysis of isolates to create less pollution, high recovery, high efficiency technology processes. General pharmacological effects of Phyllanthus, in order to provide a theoretical basis for the development and application of Phyllanthus further. The issue is the major content is divided into the following sections: 1.HSCCC separate purification of the chemical composition of Phyllanthus Objective: To take advantage of high-speed countercurrent chromatography purification of the chemical composition of Phyllanthus. Methods: The target allocation in two phases, the primaries HSCCC solvent system was observed by thin layer chromatography, and then selected by analytical HSCCC solvent system: chloroform - methanol - water, and finally carried out by adjusting the speed of the preparative separation. Results: In the analysis HSCCC injection 150mg ethyl acetate fraction extract, the solvent system chloroform - methanol - water (4:3:2), a separation into 2mg of the purity of 80.7% of the target object 1. 150mg injection water part of the extract, the solvent system chloroform - methanol - water (4:3.5:2), first separation 1.8mg purity of 90.2% for the object 2. Object 1 and object 2 can be separated on a preparative HSCCC. Conclusion: The use of the solvent system of ethyl acetate and water extraction from Phyllanthus part of the one-time separation of the desired goals. Phyllanthus extracts in vitro inhibition test purposes: Phyllanthus extract vitro antibacterial activity and inhibitory mechanisms. Methods: Staphylococcus aureus, Escherichia coli bacteria as an object, using the punch method for the determination of the suppression of Phyllanthus water raises the duty material, 75% ethanol extract, 95% ethanol extract and 95% ethanol extract extract bacteria circle diameter water extract of Phyllanthus, 75% ethanol extract, 95% ethanol extract of the minimum inhibitory concentration dilution method. Indicator bacteria, E. coli and draw the Phyllanthus water extract of Escherichia coli growth curve, the application of scanning electron microscopy to observe the the Phyllanthus water extract of E. coli morphology. Results: Phyllanthus extract on Staphylococcus aureus, Escherichia coli have a certain inhibitory effect, the minimum inhibitory concentration against Staphylococcus aureus are 31.25mg/mL minimum inhibitory concentration of E. coli is 62.5mg/mL. Phyllanthus water extract can inhibit the E. coli split logarithmic growth phase. Scanning electron microscope, the bacteria without a feeling of fullness, deformation, rupture of some bacteria. Conclusion: Phyllanthus extract has antibacterial antibacterial effect. 3 Phyllanthus Extracts antioxidant purpose: Phyllanthus Extracts antioxidant. Methods: The aqueous extract of Phyllanthus, 75% ethanol extract, 95% ethanol extracts of the hydroxyl radical (· OH), superoxide anion (O 2 · - < / sup>) Clear and inhibition of H 2 O 2 -induced mouse red blood cell hemolysis method to study the antioxidant activity of Phyllanthus. Results: concentration of 1mg/mL, water extract of Phyllanthus hydroxyl radical (· OH), superoxide anion (O 2 · - sup>) Clear and inhibition of H 2 O 2 -induced mouse red blood cell hemolysis strongest, 90.9%, 62.5%, 20.5%; 75% ethanol extract is 35.6%, 40.2 %, 16.7%; 95% ethanol extract was 48.4%, 21.3%, 17.8%. Conclusion: Phyllanthus extracts in vitro anti-oxidation. Phyllanthus extract objects outside the hepatoprotective effect Objective: To study Phyllanthus extract objects outside the hepatoprotective effect. Methods: The in vitro culture method for tilapia primary hepatocytes with CCl 4 on liver cells in vitro injury model to study the the Phyllanthus water extract, 75% ethanol extract, 95% The ethanol extract on ALT (Alanine aminitransperase, ALT), aspartate aminotransferase (Aspartate aminotransferase, AST) activity of. Results: dosing group alanine aminotransferase, aspartate aminotransferase activity was significantly lower than the injury group, the difference was significant (p <0.01). Conclusion: Phyllanthus extract has significant hepatoprotective effect in vitro.
