Yolk is shrimp nauplii embryonic and early development of the main source of nutrition. Vitellin oviparous female animals of all the major components of yolk. Decapod crustaceans Vitellin (vitellin, Vn) is a high molecular weight of fat - sugar - carotenoid proteins. Vitellin is its precursor - Vg (vitellogenin, Vg) are released into the hemolymph of oocytes absorption process (primarily glycosylation) is formed. Vitellogenin synthesis positions due to the different types and different. Crustacean ovary, pancreas, liver and adipose tissue and is likely to be the site of synthesis of vitellogenin, and most insects are mainly rely on fat body synthesis. In recent years, research on Chinese shrimp showed that: ovarian and liver pancreas are part of vitellogenin synthesis, but the two Chinese shrimp ovarian tissue at different developmental stages of vitellogenin synthesis, little quantitative research. In this study, a high sensitivity and specificity of real-time PCR method for the determination of the female Chinese shrimp in ovarian development in five different periods, ovarian and liver pancreas vitellogenin mRNA relative expression levels. Quantitative analysis of the ovaries and hepatopancreas two organizations in the process of ovarian development vitellogenin mRNA expression differences. Results of this study to elucidate the Chinese shrimp vitellogenin synthesis process sources and significance, and thus for the artificial breeding production practices provide the basis for a theoretical basis. Through the observation of Chinese shrimp anatomy, will be divided into anterior lobe of ovarian, middle, side lobe, posterior lobe of the front and rear of the posterior lobe of five parts. The results shows that: ovarian development can be divided into five stages: oogonia proliferation phase, pre-vitellogenesis, vitellogenesis primary and secondary vitellogenesis and resolution phase. Most sampling shrimp ovaries 5 different parts of the egg characteristics similar to ovarian development in the same period, a few leaves back after ovarian development in other four parts of a slow period. The results showed that: in the five stages of ovary, pancreas, ovary and liver in both vitellogenin mRNA expression, and further proof of the ovary and hepatopancreas are sites of vitellogenin synthesis. In the ovary, from the proliferation of oogonia to primary vitellogenesis, vitellogenin mRNA expression of ovarian development with an upward trend, original expressions were: 0.04,0.23 and 1.03, while in secondary vitellogenesis yolk fibrinogen mRNA expression decreased sharply to 0.16, regression of expression and significantly increased to 1.00. Primary vitellogenesis and regression of vitellogenin mRNA levels are relatively high, secondary vitellogenesis and vitellogenesis lower than the previous expression. In hepatopancreas, from the proliferation of oogonia to secondary vitellogenesis, with the development of ovaries vitellogenin mRNA levels rise, original expressions were: 0.01,0.07,0.19 and 1.29. Resolution phase of vitellogenin mRNA expression was reduced to 0.74. Secondary vitellogenesis vitellogenin mRNA expression of the highest resolution phase, followed by the proliferation of oogonia and early vitellogenesis vitellogenin mRNA expression is very low. Ovary and hepatopancreas expression comparison shows that in addition to secondary vitellogenesis, the other four periods, ovarian vitellogenin mRNA expression in the liver and pancreas more than the amount. Oogonia proliferation phase, pre-vitellogenesis primary vitellogenesis, subsided in ovarian vitellogenin mRNA levels were four times hepatopancreas, 3.2 times, 5.4 times and 1.3 times; while in secondary vitellogenesis liver pancreas expression was significantly higher than ovarian high, the former is 8.2 times the latter. In both organizations mutually complementary role, from the primary to the resolution phase of vitellogenesis, ovarian and liver pancreas overall vitellogenin mRNA levels are maintained at a high level.
