Lactic acid bacteria are normal flora of the genital tract, by generating H 2 O 2 , lactic acid, acetic acid and other substances, inhibit the growth and proliferation of pathogenic bacteria, the body's reproductive tract micro The ecological regulation plays a vital role. It has been proved, the lactobacillus can effective prevention and treatment of bovine endometritis. Lactic acid bacteria on the adhesion properties of the epithelial cells of the body is one of the important indicators as probiotics. By in vivo or in vitro adhesion test screening probiotic candidate strains, due to the limitations of in vivo studies in vitro studies based on the cellular level to become the main mode of the present study. At the same time, due to the restrictions of the appropriate cell model, little research on lactic acid bacteria adhesion of endometrial epithelial cells. Therefore, the lactic acid bacteria endometrial epithelial cells, further research and development, application of lactobacillus reproductive tract bacterial infections have potential value. This experiment Goat endometrial epithelial cells immortalized by transfection with hTERT enzyme as an in vitro model using Gram staining and plate count method to study the characteristics of lactic acid bacteria adhesion of of goat endometrial epithelial cells and its effect on pathogenic the adhesion of bacteria antagonism, and to Lactobacillus acidophilus adhesion ingredients preliminary identification, and obtained the following results: 1, our laboratory from healthy cows utero screening Lactobacillus acidophilus, Enterococcus faecalis, cecal enterococci womb epithelial cells showed a strong adhesion properties, adhesion of the number of bacteria were 87.00 ± 15.00 / cell 45.33 ± 7.506 / cells, 35.33 ± 6.110 / cell. Enterococcus faecalis and cecal enterococci adhesion ability of the difference was not significant (P gt; 0.05), significantly lower than its adhesion ability of Lactobacillus acidophilus (P lt; 0.05). 2, cow endometritis common pathogens endometrial cells with strong adhesion characteristics, including pathogenic E. coli C 83903 adhesion rate of 38.33%, pathogenic E. coli ATCC 25922 adhesion rate 65.42%, the adhesion of pathogenic Staphylococcus aureus was 86.18%. Three lactic acid bacteria Lactobacillus acidophilus, Enterococcus faecalis, cecal enterococci adhesion antagonism of pathogens pathogen adhesion rate decreased significantly (P lt; 0.01). Different lactic acid bacteria on the adhesion of pathogenic bacteria antagonism difference not statistically significant (P gt; 0.05). Escherichia coli adhesion of Lactobacillus acidophilus antagonism strongest adhesion decreased from 38.33% to 9.95%, ATCC 25922 adhesion decreased from 65.42% to 8.27% C 83903. Enterococcus faecalis and cecal enterococci Staphylococcus aureus adhesion rate from 86.18% down to 8.71%, 8.63%. 3, of goat endometrial epithelial cell growth status affect bacterial adhesion, where the largest number of bacteria, 97 / cell culture 48 h cell adhesion. Lactobacillus acidophilus has a concentration and time-dependent adhesion of endometrial epithelial cells and incubation time of 30-120 min, the adhesion of the bacteria number was significantly increased and stabilized after 120 min. As the bacterial concentration increases, the number of bacterial adhesion increased bacterial concentration of 10 9 sup>. -10 10 sup> CFU / mL, the adhesion of the bacteria count of approximately 82.33 ± 2.333 / cell; With the increase of pH, significantly reduced the number of adhesion of bacteria, wherein the pH is 4-5, the number of bacterial adhesion up to approximately 91.67 ± 1.764 / cell; lactic acid bacteria in the stability of the adhesion of the bacteria number up to $ 93.33 ± 5.686 / cell. 4, when using trypsin, lithium chloride, phenol, periodate treatment acidophilus are acidophilus adhesion capacity is significantly reduced. Showed acidophilus a variety of components involved in the adhesion process, wherein addicted played an important role in adhesion of the surface protein of the lactobacilli. 5, is extracted by the lithium chloride method acidophilus cell wall surface protein, SDS-PAGE detection, as the main surface of the size of the protein with a molecular weight of approximately 48 ku protein acidophilus. Surface protein adhesion inhibition test surface protein, strong inhibition of adhesion of E. coli to of goat endometrial epithelial cells. With the rise of the surface protein concentration, the adhesion of E. coli to of goat endometrial epithelial cells was significantly lower (p lt; 0.05), its adhesion inhibition in a dose-dependent manner.
