Abstract

Cysteamine (CS), Avian broiler targeted CS on growth performance and digestive function added to the diets of the study, and explore its mechanism. The study is divided into two parts: the first part of the study diets supplemented with different doses CS on growth performance and digestive function, serum metabolic parameters (test). The second part of the study diet, add the appropriate dose of CS on broiler growth performance of different growth stages, intestinal villi morphology, antioxidant function, serum somatostatin (SS) and insulin-like growth factor - Ⅰ (IGF-Ⅰ) content impact (test), while Research CS hypothalamus of broiler SS gene expression in the glandular stomach, duodenum and liver (test). The first part of the select 600 Yu-day-old Avian cock, were randomly divided into five groups, each group set up in each of four replicates of 30 birds, the control group was fed the basal diet, the test group were added 60,90,120,150 mg / kgCS. Each test in the 21-day and 42-day-old chicken fasting weighed and measured average daily gain (ADG) and feed conversion ratio; each group were randomly selected 12 chicken blood collection, serum metabolic parameters, and the subsequent slaughter collection pancreatic and duodenal contents, the determination of the activity of the protease, amylase and lipase. The main findings are as follows from 1.1-to 21-day stage, diets supplemented with 60 mg / kgCS can significantly improve broiler ADG add 120mg/kg, and 150mg/kgCS allows ADG decreased significantly; 22-42 day-old stage, add 90mg the / kgCS allows ADG significantly improve, and Add 150mg/kgCS so ADG decreased significantly in the broiler growth stages 150mg/kgCS group ADG and feed conversion rate was significantly lower than other groups. Tip: add CS broiler diets can improve growth performance, but high doses of CS to inhibit the growth of broiler; 1-21 days old diet add 60mg/kgCS ,22-42-day-old to add 90mg/kg CS the appropriate dose. 2.1-21 days old stage added to the diet the group pancreas 50mg/kgCS and duodenal contents protease, amylase and lipase activity significant the the higher, 120mg/kgCS and 150mg/kgCS group. 22-42 day-old stage, add 90mg/kgCS, pancreas and duodenum protease and amylase activity increased significantly compared with the control group, add 150mg/kgCS the pancreas and duodenum contents protease activity was significantly decreased, the test results suggest The digestive activity: diet supplemented with CS has a correlation with the growth performance. 1-21 days of age, stage, the diet supplemented with 60mg/kgCS significantly increased serum total protein and alkaline phosphatase, and lower serum urea nitrogen the added 150mg/kgCS significantly reduce serum total protein and alkaline phosphatase content. 22-42 day old stage, adding 60mg/kgCS, 90mg/kgCS and 120mg/kgCS serum total protein content can be significantly increased, while adding 150mg/kgCS significantly reduce the total protein. 1-21 days of age, stage, diets supplemented with different doses of CS can significantly improve broiler T3 and T4 levels, dietary supplementation 120mg/kg and 150mg/kgCS allows the gastrin levels significantly increased. 22-42 day-old stage, add 90mg/kg, of 120mg/kg and 150mg/kgCS allows serum T4 level was significantly higher than that of the control group. The serum gastrin Add 150mg/kgCS enables improved significantly. Tip: diet supplemented with CS may increase serum metabolic hormones. Select 472 birds a day-old Avian cock for the second part, were randomly divided into 2 groups, each group of eight repeat experiment was conducted in two stages in 1-21 days and 22-42 days of age, the control group was fed the basal diet test diets add the appropriate dose of CS (1-21 days old the stage add 60mg/kg ,22-42-day-old stage to add 90mg/kg) 1,7,21,28,35 and 42 days of age, each the test chicken fasting weighed to determine average daily gain, duodenum, jejunum, ileum samples collected, determination of the morphology of the intestinal villi, blood samples were collected and measured antioxidant function and serum in SS content of IGF-I, while removing the hypothalamus , glandular stomach, duodenum and liver samples, SS gene expression in different tissues was determined by semi-quantitative RT-PCR method, the main findings are as follows: 1. diets supplemented with the appropriate dose of CS can significantly improve broiler 21,28 35 and 42 days of age, body weight, and stage 14-21 days old and 21-28 days old stage trials chicken ADG than the control group increased by 9.48% (P lt; 0.05) and 8.42% (P lt; 0.05) . The test results suggest that: diets supplemented with appropriate doses of CS to promote the growth of broiler effect is more pronounced in the 14-28 days of age. Stage in the 21-day-old diets supplemented with CS significantly reduced duodenal villus height and crypt depth of the jejunum, ileum villus height and crypt depth had no significant effect; 42-day-old stage, add the CS duodenum, jejunum and ileum villus height were not significantly affected. CS can significantly increase the 7-day-old, 14-day-old and 21-day-old broiler serum glutathione peroxidase activity and increased the total antioxidant capacity of 7-day-old broiler. Add CS in 14, 21 and 28 days of age, the diet can significantly reduce the level of broiler serum SS significantly improve the level of serum IGF-Ⅰ. . Diets supplemented with CS no significant effect on the 7-28 day-old stage hypothalamus SSmRNA expression significantly reduced the relative expression level hypothalamus SSmRNA's at 35 days of age, was significantly higher at 42 days of age; Add CS can significantly reduce the 28-day-old broiler proventriculus SS mRNA expression levels; CS can promote growth during broiler the duodenum SSmRNA of expression in the 21-day, 28-day-old reached a significant level. CS SS gene expression in the liver is inhibited significantly at 35 days of age reduce liver SSmRNA expression. Results Tip: add CS stage 1-14 day-old broiler SS gene expression in the diet had no significant effect, while the 22-42 day-old stage SS gene expression significantly affected, diets CS SS gene expression tissue growth stage specificity. In summary: Add CS broiler diets promote growth, improve the digestive enzyme activities, but high doses of CS to inhibit the growth and reduce the activity of digestive enzymes. 1-21 days old stage 60mg/kg ,22-42-day-old stage 90mg/kg diet appropriate CS dosage. Add CS can increase the serum metabolic hormone level. Add the appropriate dose CS diet can increase the growth of broiler pre antioxidant capacity and serum level of IGF-Ⅰ, lower serum level of SS. Add a CS 1-14 day-old stage broiler SS gene expression, while the 22-42 day-old stage SS gene expression had a significant effect, the organization and growth stage-specific diets CS to SS gene expression . The test results suggest that diets supplemented with CS promote broiler growth is mainly achieved by increasing the activities of digestive enzymes and the level of IGF-Ⅰ, reduce the level of serum SS.
