Abstract


Cucumber (Cucumis sativus L.) caused by the Cuban false downy mildew (Pseudoperonospora cubensis) infection of downy mildew infection in the cultivation and production often cause serious economic losses. Plant chitinase hydrolysis of fungal cell wall chitin to withstand pathogen infection, an important class of pathogenesis-related proteins (of defecation and-related proteins, PRP). Plants chitinase are mainly distributed in the cell gap and vacuoles, intercellular spaces First-induced accumulation of chitinase often pathogen infection. Therefore, the accumulation of chitinase research intercellular space variation of significance to reveal the mechanism of the reaction of the plant pathogen defense. 1 In this paper, using different varieties of cucumber downy mildew, inoculated with downy mildew cucumber seedling leaves intercellular space fluid accumulation of chitinase induced dynamic changes and inoculation on seedling growth. The results show that: Compared with easily susceptible cultivar Changchun Mici \in the first two days after inoculation, reached the peak, and the high activity is maintained up to 7 days; observed by SDS-PAGE electrophoresis, one kind 27kDa chitinase in the rapid accumulation of the first two days after inoculation, and after inoculation 4 7 days showed higher expression levels; confirmed by Western blotting was the result of the above chitinase expression change. After inoculation, disease-resistant cucumber leaves ethylene production increased rapidly, and the large range of susceptible varieties increase ethylene production in late inoculation. In addition, compared with the control, 4,7 days after the inoculation treatment susceptible cucumber seedling fresh weight, dry matter accumulation of a significant reduction in the volume appear seven days after inoculation, the growth of resistant varieties indicators to be significantly higher than the susceptible disease varieties. The above results show that the intercellular spaces chitinase and cucumber downy mildew resistance is closely related. Ethylene is considered a signaling molecule reactions plant resistance to pathogen infection, and plant disease resistance response synthetic PRP closely related. Ethephon chemical compounds that can be the plants quickly absorbed and released ethylene. In this paper, the 4th Jinchun cucumber as test materials, cucumber seedling leaves sprayed with different concentrations of ethephon on the intercellular fluid chitinase induced accumulation of dynamic change and on seedling growth. The results show that: cucumber leaves compared with the control, Ethrel intercellular space increased protein content; chitinase activity levels of different amplitude increases with the concentration of Ethrel, where 0.02% ethephon chitin chitinase induced strongest; observed by SDS-PAGE electrophoresis, ethephon to 0.02% after 1 day, the accumulation of one kind 27kDa chitinase in the intercellular spaces, and in the subsequent sampling time significant expression. Western blotting analysis further confirmed the above results. In addition, compared with the control, 0.01% and 0.02% after ethephon 7 days, cucumber seedling height increments significantly reduced, stem diameter increment of 0.02%, a significant increase. The above results show that ethephon can induce the cucumber intracellular gap chitinase generated accumulation induced effect with cucumber feelings directly related to the ethylene concentration. This study shows that, downy mildew inoculation or can be induced by Ethrel the cucumber leaf intercellular spaces 27kDa chitinase accumulation vaccination processing is closely related to the 27kDa chitinase induced release of ethylene and cucumber leaves.
