The purpose of the model rats kindled prepared on the basis of conduct Erdosteine intervention rat hippocampal superoxide dismutase (SOD) activity and malondialdehyde (MDA), and changes in IL-1β, IL-6, TNF-α and MMP-2 levels and to explore Erdosteine influence the pathogenesis of epilepsy for epilepsy provides a potentially effective new type of single-agent multi-target drug benefit for patients. Method 1. Experimental animals and grouping: Healthy adult male Wistar rats were randomly divided into three groups: group Pentylenetetrazol (n = 10), Erdosteine intervention group (n = 10) and normal control group (n = 10 ). (2) Preparation and intervention models of epilepsy: A Diehl kindling model making methods, PTZ group and intervention group Erdosteine intraperitoneal injection of PTZ kindling model of epilepsy 50mg/Kg production, Erdosteine intervention group was ignited ago by gavage Erdosteine 50mg/Kg intervention treatment. 3 drawn and experimental determination: All models meet standards lit experimental animals were decapitated, the brain, the hippocampus rapidly removed by chemical colorimetric assay SOD activity in hippocampus and MDA content; rats were observed by HE staining hippocampal neurons injury; using double antibody sandwich ELISA assay cytokines IL-1β, IL-6 and TNF-α levels change; immunohistochemical staining using the MMP-2 positive cells. 4 Statistical analysis: The resulting count data as mean ± standard deviation (x ± s) description and level data are analyzed using SPSS17.0 statistical software. Results 1.HE staining: control group rat hippocampal neurons tightly packed, sharp edges, transparent cytoplasm, nuclear morphology was normal, no necrotic neurons. PTZ hippocampal neurons arranged scattered, reducing the number of the cell body shrinkage, cytoplasmic and nuclear condensation red dye, a large number of necrotic neurons; Erdosteine intervention group scattered in hippocampal neuronal necrosis element between the two groups was significant difference (p lt; 0.01). 2.MMP-2 immunoreactive cells were mainly distributed in the hippocampus vertebral cell layer and granule cell layer, but also in the molecular layer. Normal group and intervention group Erdosteine staining intensity compared with the model group decreased significantly (p lt; 0.05). 3 PTZ group and intervention group Erdosteine SOD activity in hippocampus compared with the normal group were lower, but the PTZ group than in the intervention group Erdosteine decreased more significantly (p lt; 0.01). Compared with normal group, MDA Pentylenetetrazole group was significantly higher (p lt; 0.01), Erdosteine intervention group than in the group decreased significantly PTZ (p lt; 0.01). 4 PTZ group and intervention group Erdosteine hippocampal IL-1, IL-6 and TNF-α levels compared with the normal group were significantly increased (p lt; 0.05), but the model group increased more significantly (p lt; 0.01). Conclusion seizures associated with oxidative stress, cytokines and neuronal damage. Erdosteine suppress epileptic rat neuronal damage, scavenging free radicals and reduce the role of cytokine production, and can inhibit the activation of MMP-2, has a protective effect on the brain tissue.
