Validation on the Interaction between Jumping Translocation Breakpoint Protein and Envelope Protein of Hepatitis B Virus

Objective: This study mammalian cell two hybrid , co-immunoprecipitation and GST pull -down and other methods to further confirm the translocation of HBV envelope protein (HBsAg) and fracture jump (Jumping Translocation Breakpoint, JTB) the interaction between the protein . Method: using the the Stratagene mammalian cells produced two-hybrid system for verification . PCMV -BD - JTB pCMV -AD - full S plasmid and reporter plasmid pFR-Luc , was transfected into 293FT cells to confirm HBsAg and JTB protein interactions . BD-JTB collection 293FT cells expressing S protein of the protein and AD- full two-way co-immunoprecipitation experiments further confirmed , and JTB-GST pull-down JTB protein main S - protein interactions . Results : The two-hybrid mammalian cells prompts HBsAg and JTB protein , luciferase expression in transfected group excited above the positive control group , suggesting that there is interaction between the two ; verify the results of the two-way co-immunoprecipitation and GST pull-down method prompts the presence of HBsAg and JTB protein interactions, and the combination of the two sites may be located in the main S binding domain . Conclusion: HBsAg and JTB protein exists interaction, the binding site is located in the main S binding domain .
