Abstract


Marine life in genomics and proteomics research increasingly rapid development of the urgent need to introduce bioinformatics methods and means to explain the massive data, crack repositories of biological information, the information prompted for the experimental study. This thesis is to build a Linux-based operating system cluster computer environments and J2EE distributed computing technology, bioinformatics data analysis platform-the BLC (Bioinformatics Linux Cluster) system. The system includes a Blast, Pfam, Phrap, Interpro bioinformatics software, can easily annotate molecular sequence data, analysis and computation. On this basis, the integration team has sequence resources, the formation of the Chinese economy class marine organisms genome website. Chinese shrimp and clams with immune and growth and development in order to screen the function of genes, has been constructed from the shrimp blood were measured, the eyestalk and ovarian tissue cDNA library in 2371, 1024 and 1172 EST. And cDNA library constructed clam larvae at different developmental stages, from 1060 EST sequences are measured in the clam shell top stage larvae cDNA library. The bioinformatics platform constructed in this study to analyze, compare, and annotate the shrimp, clams EST. And obtained from the Chinese shrimp blood cDNA library of 971 unique genes discovered 112 new genes associated immune. Further screening identified a total of antimicrobial peptides play an important role in shrimp immune system, the clotting protein, cell signaling, chaperone protein and phenol oxidase system in five major categories of a total of 39 unique genes. Found larvae of the clam shell top 576 unique genes, including 150 with the growth and development of the growth and structure of genes, metabolic enzymes, immune-related genes, regulatory genes important function of gene sequences suggest. Part of the bioinformatics analysis results and cloned a 2370 bp shrimp cell adhesion protein gene length cDNA sequence encoding 684 amino acids, the mature peptide theory pI 7.98, molecular weight 74.58 KDa is a cationic polypeptide. The sequence has a peroxidase function domain (domain) and an integrin binding motif (motif), inferred that the gene has peroxidase activity and cell adhesion. Gram-positive and Gram-negative bacteria using real-time quantitative PCR analysis inactivated mixture stimulus shrimp cell adhesion protein gene expression profiling results show that its main storage in blood cells was constitutive expression of prawn important immune response one of the factors having important physiological functions in the process of immune defense.
