Abstract


Yersinia pestis(Yersinina pestis, hereinafter referred to as Yersinia pestis) has this in history triggered a worldwide three times plague outbreak, brought to mankind the unprecedented disaster. Plague has been able to so rampant buffeting the whole world, mainly because of the plague as a natural focal disease with its fast spreading rate, sudden illness is acute and unpredictable, the mortality rate was high and highly contagious characterized Group A potent infectious diseases. History has never been any other like the plague, a disease to humans has brought so much disaster, such painful memories left to humans. These methods are not suitable for fine phylogenetic analysis. The resolution of the oriental strains of plague were classified using a variety of genetic markers, can be avoided using a single genetic marker or partial results.1.Yersinia pestis as a relatively "young" and the lack of recombinant pathogenic bacteria, single nucleotide polymorphisms (Single Nucleotide polymorphisms, SNP), can be used as a very reliable genotyping and phylogenetic analysis of the indicators. This study established the Y. pestis SNP genotyping technology based Lumienx x-TAG microspheres (liquid suspension array) using multiplex PCR amplification of Y. pestis18SNP loci fragments, the amplified product of complete multiplex the ASPE (Allele the Specific primer Extension) multi-SNP detection reaction and Luminex technology, the base characteristics of the tested strains of the18SNP loci can be detected by a reaction. Use this method to analyze the20plague oriental strains can be divided into seven genotypes according to the characteristics of the18SNP loci, this method has the interpretation of simple, rapid, high throughput, low cost, the SNP in the pathogen typing has broad application prospects.2. Preliminary study of the origin of the Yersinia pestis in China Oriental.The international community generally agreed that the third plague pandemic originated in Yunnan, China, oriental strains of Yersinia pestis originated in Yunnan. But our early research results with a point in some respects does not match that of Yunnan oriental strains are relatively "young" strain, while the oldest oriental strains isolated from Fujian. In this study, the use of the Luminex technology platform detection Yunnan oriental strains of Yunnan oriental strain typing results of a preliminary study, genotyping, a total of193plague Oriental strains into13genotypes, a, b, c, d, e, f, g, and h, i, j, k,1, m, respectively.53SNP information provides important data in the late period to trace the origin of the oriental plague bacteria.
