Abstract

Aging and longevity is one of the most difficult and complex issues faced by the Human Sciences Research, aging is a result of the role of the two genes and the external environment. Which genes determine cellular senescence, the aging of tissues and organs, a key factor within the overall aging of the system is aging and body. How to modulate the activity state of senescence-related genes, from aging the source initiative intervention, start to slow down or reverse aging and the development process is the key to this research. This article through the collation and analysis of the nematode longevity gene has been confirmed, these genes to construct the nematode longevity partial gene network. From the point of view of the gene network research can fully reflect the internal laws of the phenomena of life itself is better than in the past to study a single gene in isolation. Were selected and C. elegans as a model organism, total choice of two programs feeding nematodes. Respectively, for different types of lactic acid bacteria and a branched-chain amino acids and combinations thereof. The different nutrients feeding nematodes mode and filter out the optimal combination enables nematode longevity. Using real-time quantitative PCR using the optimal combination feeding nematodes in multiple stages of growth of the gene expression changes, the genes in the nematode partial longevity gene network selected genes were constructed. More intuitive description of the impact of the external environment of the longevity network, the interaction between the various gene can also have a preliminary understanding. Of course, for the study of complex biological traits not yet fixed research strategies and methods, many environmental factors and sensitive gene interactions can have an impact on biological traits occur, this process will be a dynamic cycle. The test results show that the part of the lactic acid bacteria in the NGM plate can not grow well, which gives difficulties brought to the survival and metastasis of nematodes. In addition to these bacteria go directly to the test. Finalized a total of 11 of the feeding nematode strains, were No. 1, No. 5, No. 8, 1936 No. ,119-10 ,6-3-1 ,6-3-2 ,7-3-1 No. ,7-3-2 ,6-4 ,7-4. 11 lactic acid bacteria in the test, not all of lactic acid bacteria life has only four pairs of the nematode's life had an impact. Can be reduced in 1936 ,6-3-2 ,6-4 ,7-4, respectively, and all lactobacillus species of nematode life impact is not reflected in the of nematodes maximum life extension of, but rather reflects nematodes pre mortality. Second nutrients for the branched-chain amino acids, nutrients directly added to the culture medium, still uses the E.coli OP50 feeding nematodes. Use three kinds of amino acid altogether, respectively, valine, leucine, isoleucine, each amino acid with a concentration of 3 tests, respectively, for a 0.1-fold concentration, 1-fold concentration, 10-fold concentration. Then choose the optimal concentration of each amino acid combination trials. The test results show that the longevity of the effect of the combination of branched-chain amino acids and the nematode life is better than a single branched chain amino acids. Nutrients and combinations of all test trials, the effect of 0.1 times valine. And this effect is not only reflected in the of nematodes maximum life extension of, but also the performance of the mortality of the entire life cycle of the nematode reduction. Valine metabolism around 14 genes by real-time quantitative PCR analysis of the expression level of, the results show that SPTL-2 CYP-33E1 C16A3.10, ODC-1, C2.8H8.11, ALH-13 6 The increase in the amount of gene expression. unc-25 gene expression level decreased. C05D2.3 genes at different stages have different effects, the decline in the early expression levels, improve expression levels in the medium term. From different genetic changes differences, we can further infer the function of the gene associated with longevity and longevity. Can also speculate that those genes has not been reported longevity sensitive genes, lay the foundation for the follow-up study. Build the longevity gene expression database, can the the nematode longevity detection data support.
