Abstract

In this thesis, the sequential injection (SI) technique coupled with chemiluminescence (CL) detection was developed for the determination of Carbaryl and Hemoglobin (Hb) and satisfactory results were obtained. The first section of the thesis introduced the brief history and classification of pesticide, the pollution to environment and the detection technique of pesticide residue. And the section especially reviewed the application of chemiluminescence analytic technique in pesticide residue detection. In the second section, a procedure for the determination of Carbaryl by sequential injection analysis coupled with chemiluminescence was described, which was based on the greatly increasing of chemiluminescence intensity of Carbaryl’s oxidation with Ce(IV) in a nitric acid medium containing Rhodamine 6G as sensitizer. The sampling sequence of reagents and sample, the concentration and volume of reagents, the flow rate and so on were optimized by univariate method. Under the optimized condition, the chemiluminescence intensity was linear with the concentration of Carbaryl over the range 1.0×10-4-2.0×10-1mg/L, the detection limit (3σ) was 3.0×10-5 mg/L and the relative standard deviation for eleven repeated measurements for 0.10 mg/L Carbaryl was 0.44%, and sample throughput was 132 samples per hour. The method was applied to the determination of Carbaryl in environment water samples as well as in grain samples and the recoveries were over 91.3-107%.In the third section, a SI-CL method was developed for the determination of Hemoglobin. It was based on the intense chemiluminescence generated by the oxidation-reduction reaction of Luminol and Potassium ferricyanide under the concerted catalysis of Hemoglobin and hexadecyl trimethyl ammonium bromide (CTMAB) in alkaline medium. The chemiluminescence intensity was linear with the concentration of Hemoglobin over the range 1.0×10-2-10.0 mg/L, the detection limit (3σ) was 3.0×10-3 mg/L and the relative standard deviation for eleven repeated measurements for 0.50 mg/L Hemoglobin was 0.54%, and the sampling frequency was 136 samples per hour. The method was applied to the determination of Hemoglobin in human serum and the recoveries were over the range 94.7-105%.In the last section, a summary for content of the experimentation was described.
