Abstract


Objective1Evaluation the bufei jianpi recipe, bufei yishen recipe and yiqi zishen rec-ipe on the therapeutic effect of COPD and its late effects arising, and to in-vestigate its characteristics.2Explore the mechanism of bufei jianpi recipe, bufei yishen recipe and yiqi zishen recipe to COPD and reveal the mechanisms of late effects.MethodsThe rats were randomly divided into control group, model group, bufei jianpi group, bufei yishen group, yiqi zishen group and aminophyline grou-p.Combined with the repeated smoke inhalations and bacterial infections of the method to production the stable COPD rat model.From the nineth week, we must discern supply drug to the rats, in the model prepared por the twen-ty week, half the animals were sacrificed, the remaining animals breed nor-mally to32weeks, Put to death. We can check out the lung function(TV, PEF, EF50), peripheral blood and bronchoalveolar lavage fluid leukocyte count and neutrophil percentage, lung pathology(bronchial wall thickness, degree of bronchial stenosis, pulmonary vascular wall thickness, unit area of alveolar count and alveolar diameter), and the cytokine in the blood se-rum and BALF(IL-1β, IL-6, IL-8, IL-10, TNF-α and sTNF).Results1General state of healthCompared with the control group, in the model group,the rat’s body weight is magersucht, the spirit is fatigue, the activities is reduced, the food and water intake is decreased, the stool is thin pond and the bedding is moi-st etc. The Bufei Jianpi recipe, Bufei Yisheng recipe, Yiqi Zishen recipe an- d aminophylline recipe can all improve the change, especially in rats treated with Bufei Jianpi recipe and Bufei Yisheng recipe. Weight of rats in each group increased with increasing weeks of age. Body weight of rats in model group than in the control group of slow growth trend(P>0.05). Bufei jianpi group, bufei yishen group, yiqi zishen group and aminophylline group from the first12weeks of weight compared with the model group to accelerate growth trend, but among the groups was no significant statistical difference (P>0.05).2Lung functionlnThe TV, PEF, EF50level of model group from the first4weeks was significantly lower than the control group (P<0.05orP<0.01). Compared with model group, bufei jianpi group, bufei yishen group and yiqi zishen group from the16th week clearly increase the level of PEF and EF50(P <0.05or P<0.01), TV values increased significantly from20weeks on-wards(P<0.05orP<0.01). Aminophylline group only at20weeks and28weeks was significantly higher of PEF(P<0.05). In the32weeks observ-ation period, the level of TV, PEF, EF50between bufei jianpi group, bufei yishen group, yiqi zishen group and aminophylline group was no significant difference(P>0.05).3WBC count and neutrophil percentage of Peripheral bloodAt the end of the20and the32week, model group counts in peripheral blood WBC and neutrophil ratios tended to increase compared with the control group, but no significant statistical difference(P>0.05).4WBC count and neutrophil percentage of BALFAt the end of the20and the32week, the WBC count of model group were significantly higher than control group(P<0.01); compared with model group, bufei jianpi group, bufei yishen group and yiqi zishen group were significantly lower(P<0.01). Compared with model group, aminophylline group were significantly lower at20week(P<0.05), no significant difference at32week(P>0.05). Compared with the aminophylline group, bufei jianpi group was significantly lower at20week and32week(P<0.05orP<0.01); bufei yishen group and yiqi zishen group were significantly lower at32week(P<0.05). There is no significant difference between bufei jianpi group, bufei yishen group and yiqi zishen group. Neutrophil percentage between the groups was no significant difference(P>0.05).5Lung pathologyAt the end of the20and32week, the bronchial wall thickness, degree of bronchial stenosis, pulmonary vascular wall thickness and alveolar diameter were significantly higher than the control group(P<0.01), the unit area of alveolar count was significantly lower(P<0.01). Compared with model group, the pathological changes of bufei jianpi group, bufei yishen group, yiqi zishen group and aminophylline group were significantly reduced(P<0.05orP<0.01). At the end of32week, compared with aminophylline group, bufei jianpi group, bufei yishen group and yiqi zishen group are significantly lower at bronchial wall thickness, pulmonary vascular wall thickness and alveolar diameter(P<0.01);there is no significant difference bewteen bufei jianpi group, bufei yishen group and yiqi zishen group(P>0.05).6Cytokines of serumAt the end of20week, the IL-1β, IL-6, IL-8, IL-10, TNF-α, soluble TNF receptor level of model group were significantly higher than control group(P<0.01). The cytokine levels of bufei jianpi group, bufei yishen group and yiqi zishen group were significantly lower than model group (P<0.01).Among them, the IL-1β, IL-10, TNF-α levels of yiqi zishen group were significantly lower than bufei yishen group(P<0.01); the IL-6, TNF-α levels of bufei jianpi group were significantly lower than bufei yishen group and yiqi zishen group(P<0.05orP<0.01); the IL-8level of yiqi zishen group was significantly lower compared with bufei jianpi group and bufei yishen group(P<0.05); the soluble TNF receptor level of bufei jianpi group and yiqi zishen group were significantly lower than bufei yishen group (P<0.01).The IL-1β, IL-6, IL-8, IL-10, TNF-α, soluble TNF receptor levels of bufei jianpi group, bufei yishen group and yiqi zishen group were sign-ificantly lower than those of aminophylline(P<0.01). At the end of32week, the IL-1β, IL-8, IL-10, TNF-α levels were still significantly lower than model group(P<0.05orP<0.01); the IL-6, soluble TNF receptor levels of bufei jianpi group and bufei yishen group were significantly decreased compared with yiqi zishen group and aminophylline group(P<0.01); the IL-8level of bufei jianpi group was significantly lower than yiqi zishen group(P<0.05); the IL-10, TNF-α levels of bufei jianpi group and bufei yishen group were significantly lower than yiqi zishen group(P<0.05or P<0.01).7Cytokines of BALFAt the end of20and32week, the IL-8, IL-10, TNF-α levels of model group were significantly higher than control group (P<0.01); these cytokine levels of bufei jianpi group, bufei yishen group and yiqi zishen group were significantly lower than model group(P<0.01). Among them, at the end of20week, the IL-8, IL-10levels of yiqi zishen group were significantly lower than bufei yishen group(P<0.05orP<0.01); the TNF-α level of bufei jianpi group was decreased significantly than bufei yishen group and yiqi zishen group(P<0.05orP<0.01); the TNF-α level of yiqi zishen group was significantly lower than bufei yishen group(P<0.01). At the end32week, the IL-8, TNF-α levels of bufei jianpi group and bufei yishen group improved significantly than yiqi zishen group(P<0.05orP<0.01); the IL-10level of bufei yishen group was significantly lower than yiqi zishen group (P<0.01). The IL-8, IL-10, TNF-α levels of bufei jianpi group, bufei yishen group and yiqi zishen group were significantly lower than those of aminop-hylline(P<0.01).8Correlation AnalysisThat is Significant positive correlation between degree of bronchial stenosis and level of serum IL-8, IL-6, IL-10, IL-1β, TNF-α, soluble TNF receptor(r=0.643,0.657,0.619,0.641,0.648,0.663, P=0.000); of BALF IL-8, IL-10, TNF-α (r=0.593,0.612,0.656, P=0.000).That is Significant positive correlation between bronchial wall thickness and level of serum IL-8, IL-6, IL-10, IL-1β, TNF-α, soluble TNF receptor(r=0.776,0.784,0.740,0.728,0.776,0.770, P=0.000); of BALF IL-8, IL-10, TNF-α(r=0.706,0.732,0.775, P=0.000).That is Significant positive correlation between pulmonary vascular wall thickness and level of serum IL-8, IL-6, IL-10, IL-1β, TNF-α, soluble TNF receptor(r=0.650,0.716,0.657,0.687,0.706,0.686, P=0.000); of BALF IL-8, IL-10, TNF-α(r=0.706,0.732,0.775, P=0.000).That is Significant negative correlation between unit area of alveolar count and level of serum IL-8, IL-6, IL-10, IL-1β, TNF-α, soluble TNF receptor(r=-0.494,-0.444,-0.490,-0.463,-0.432,-0.431, P=0.000,0.002,0.000,0.001,0.002,0.002); of BALF IL-8, IL-10, TNF-α(r=-0.480,-0.484,-0.434, P=0.001,0.000,0.002).That is Significant positive correlation between alveolar diameter and level of serum IL-8, IL-6, IL-10, IL-1β, TNF-α, soluble TNF receptor(r=0.743,0.770,0.731,0.769,0.773,0.761, P=0.000); of BALF IL-8, IL-10, TNF-α (r=0.777,0.731,0.768, P=0.000).Conclusion1Bufei Jianpi recipes, bufei yishen recipes and yiqi zishen recipes can obviously improve the whole condition, pulmonary function and pathological changes in Rats of stable COPD, as well the remote effect.2There have close relation between pathological changes in lung tissue and and the expression of IL-8、IL-6、IL-10、IL-1β and TNF-α, it show that, inflammatory response to injury is one important mechanism about the the development of COPD.3There are good effect and remote effect of bufei Jianpi recipes, bufei yishen recipes and yiqi zishen recipes in COPD rats, the mechanism may be involved in regulating the level of inflammatory cytokines. The effect of Bufei Jianpi recipes and bufei yishen recipes are better than yiqi zishen recipes.
