Abstract


Objective:In the1990s, Belgium researchers found women appeared hematruia, waist pain, kidney dysfunction after taking Chinese medicine when they were losing weight, part of whom got the uremia disease and received dialysis treatment, finally the pathological diagnosis was renal fibrosis. This kind of renal damage in the European was called "Chinese medicine nephropathy". Since the reports of the damage of Chinese medicine were increasing gradually, domestic scholars had made lots of studies on this question, then the Chinese medicine nephropathy was identified, such as the affair of the aristolochinc acid nephropathy and long gan xie dan pills. As a long history of traditional medicine,Zexie promote water metabolism, cleanse phlegm retention and remove the hot humid of the body. A large number of clinical studies showed zexie and its compound have good curative effective on Hyperglycemia, fatty liver, immune complex type and the basement membrane type nephritis, kidney stone, high blood pressure. There were reports about its hepatotoxicity. The Chinese medicine adverse reactions and preventions recorded:ahige dose or long-term consumption of zexie can cause water-electrolyte imbalances, hematuria and acidosis. With the guide of traditional Chinese medicine theory and modern medical theory, this experiment explores the kidney toxicity of zexie and its fractions.Methods:A total of140kunming mice were divided into seven groups, each consisting of10female mice and10male mice, they are:the high dose of the aqueous extract, the low dose of the aqueous extract, the high dose of the fraction1, the low dose of fraction1, the high dose of the fraction2, the1ow dose of thef ract i on2. The normal group are given distilled water20mL. kg-1, d-1by irrigating gastric, the high and low dose of the aqueous extract mice are given13.75g. kg-1. d-1and8.25g. kg-1, d-1by irrigating gastric respectively, the high and low dose of the fraction1mice are given5.5g. kg-1. d-1and3.2g. kg-1. d-1byirrigating gastric respectively, the high andlow dose of the fraction2mice are given5.64g. kg-1, d-1and3.4g. kg-1, d-1by irrigating gastric respectively. The proc ess will take10weeks. The urine of each group mice is collected by the end of the experiments, then, the mice are fast and given water for24hours. The whole blood was collected by ophthalmectomy, the kidney are removed, part of the kidney are used for pathological microscopic observations. Results:Compared with the normal mice, the female and male mice in the high dose of fraction1group, the BUN, CR, NAG and Y-GT have significantly increased (P<0.01) while the female and male mice in the high dose of the aqueous extract group, the BUN, CR, NAG and y-GT have significantly i ncreased (P<0.05), the female and male mice in the high dose of fraction2group, the CR and NAG have significantly increased (P<0.05). The expression of BMP-7have significantly decreased (P<0.05) in the high dose of the aqueous extract group, the fraction1group and the fraction2group while the expression of MCP-1have significantly increase (P<0.05) in these three groups. For the female mice in the high dose of the fraction1group, the expression of BMP-7is lowest and the express i on of MCP-1i s h i ghest. Conclusion:1The female and male mice in the high dose of fraction1group, the BUN, CR, NAG and Y-GT have significantly increased (P<0.01) while the express i on of BMP-7is lowest and the expression of MCP-1is highest.2The high dose of fraction1has the most serious damage to kidney.
