Abstract


Acorus calamus Linn. is a member of Acorus genus, Araceae family. The dry rhizome of Acorus calamus is used as "Zangchangpu" in Triditional Chinese Medicine.In this thesis, chemical constituents from70%ethanol extract of Acorus calamus were studied. On the basis of different kinds of modern chromatography techniques (silica gel, ODS, HPLC and Sephadex LH-20) and spectroscopic methods (MS,1H-,13C-NMR),10compounds were isolated from Acorus calamus and identified as:1β,4β,7α-trihydroxyeudesmane (1), bullatantriol (2), teuclatriol (3), threo-1’,2’-dihydroxyasarone (4), erythro-1’,2’-dihydroxyasarone (5),(+)-de-4’-O-methyleudesmin (6),(+)-de-4’-O-methylmagnolin (7),(+)-eudesmin (8),(+)-magnolin (9),β-sitosterol (10). Compounds1,2,4-9were isolated from Acorus calamus for the first time. Compounds1,2,6,7,8,9were obtained from Acorus genus for the first time.Meanwhile, in vitro antidiabetic activities of the extracted fractions were investigated using3T3-L1cell line. In the adipocyte differentiation assay, compared to the solvent control group, lipid droplets in the ethyl actate and water fraction groups increased significantly, similar to the rosiglitazone positive control group. Glucose consumption of the n-butanol fraction group and rosiglitazone group increased obviously. In the ethyl acetate fraction group and70%ethanol extract group, the glucose consumption increased significantly.The antidiabetic activities of the isolated compounds were evaluated on HepG2 cell model of glucose consumption. Glucose consumptions in1μg·mL-1,5μg·mL-1, and25μg·mL-1groups of compound1,3,6,7,8increased significantly with dose-dependence (P<0.05, P<0.01or P<0.001), and all the increasing rates were greater than20%. The glucose consumption increased83.4%in the positive control group (metformin) at1mmol·L-1. The result suggests that the sesquiterpenes and lignans might be responsible for the antidiabetic activity of Acorus calamus.In conclusion, this research has provided the theoretical and experimental basis for the development and quality control of Acorus calamus.
