Abstract


Sub-health indicates series of functional changes without pathological change of human. It is also called "Uncertainly Symptom" by multiple and uncertain symptoms. A investigation of WHO indicates that there is about 5% people who are healthy in deed, 20% on disease and 75% sub-health. It seems an increase trend of sub-health in many countries and areas. Sub-health has become a latent killer which is harmful to human’s health and it is also a puzzle and hotspot in modern medicine. Therefore searching for some effective measures has become an important problem which is in dire need to be solved. To establish a kind of sub-health model is the key point to make the researches deepened at present. However there are not such models for the pathogen and mechanism of sub-health haven’t been found out yet. As animal experiment can be controlled、operated and repeated so easily and conveniently that it is one of the most important methods in modern biomedical research. Hence it is much necessary to establish the sub-health animal model for investigating the rule of occurrence、development and the methods of prevention and cure of sub-health.This experiment includes two parts. The first part is based on the diagnosis standard of sub-health of human——"no obvious evidences of organic pathology damages and positive changes of laboratory targets through systematic examination." According to etiology and symptom simulation, we adopted forced-swimming as the method to establish a rat model of sub-health. Then we estimated the model through the body temperature, the exhausted swimming time、blood、urine and tissue pathology examination. The second part is based on the theory of Chinese traditional medicine——"liver is the essence of fatigue". We observed weikang granule, a kind of herbal medicines, affecting fatigue-resistance、liver function、liver tissue pathology on the rat model of sub-health and discussed the mechanism and theory of sub-health.ChapterⅠEstablishment of a rat model of sub-healthObjective: To establish a rat model of sub-health.Method:Experiment 1: 100 rats were randomly divided into two groups: The normal control group(C) and the model group (M), 50 rats per group. The model group was given forced-swimming exercise without load 2h/d in a 120cm×60cm×60cm swimming pool filled with water (24±2℃) to a height of 30cm. The normal control group was kept fasting and water-privation during the model group’s swimming. The model groups’ exhausted swimming time was recorded before modeling and every 7 days separately. We observed the effect of fatigue caused by forced-swimming dynamically at different time plots. 10 rats of the normal control group and the model group were executed every 7days and then examined the Blood Routine Test、the Urine Routine Test、K+、Na+、CL-、Ca2+、BUN、CR、ALT、AST、TBIL、TP、ALB、G、GLU、TG、CHOL、LDH、CK. The heart、liver、spleen、lung、kidney、brain、stomach、intestine、lymph、gonad were taken pathological examination. Experiment 2: 36 rats were randomly divided into two groups: the normal control group(C) and the model group (M), 18 rats per group. Rats of the model group were forced swimming without load 2h/d in a 120cm×60cm×60cm swimming pool filled with water (24±2℃) to a height of 30cm. We observed the exhausted swimming time、ALT、AST and liver pathological examination.Experiment 3: 6 rats were forced swimming without load 2 b/d in a 120cm×60cm×60cm swimming pool filled with water(24±2℃) to a height of 30cm. Then we stopped rats swimming at day 13th and observed the exhausted swimming time during the recovery period.Results:Experiment 1:1、The common status: all rats’ body temperature maintain 37.8~38.7℃, no fever and lymphadenectasis. Compared with the normal group, rats’ activity and reactivity of the model group declined significantly.2、The exhausted swimming time: the exhausting swimming time of the model group decreased significantly (P<0.05) since the 2nd week and had a decreasing tendency but without significant difference (P>0.05) compared with it before modeling.3、Blood Routine Test: compared with the normal control group, WBC、RBC、HGB had no significant difference (P>0.05) but PLT indicated a significant difference at the 5th week (P<0.05).4、Urine Routine Test: the control group and the model group didn’t find WBC、RBC、protein in urine.5、Blood biochemistry level: compared with the normal group, K+、Na+、CL-、Ca2+、TP、ALB、G、TBIL、CR、GLU、CK、LDH of the model group have no significant difference during 5 weeks(P>0.05). There was a significant increase level of BUN from the 2nd week and lasted 5 weeks (P<0.05). The level of ALT indicated a significant increase from the 3rd week to the 5th week (P<0.05 or P<0.01) and AST also had a significant increase from the 2nd week to the 5th week (P<0.05 or P<0.01). TG and CHOL indicated significant differences between the 3rd and 5th week (P<0.05).6、The pathological examination: all rats indicated no obvious changes in heart、liver、spleen、lung、kidney、brain、stomach、intestine、lymph、gonad at the 14th day.Experiment 2:1、The exhausted swimming time: compared with each group before modeling, the exhausted swimming time at the 10th、12th day declined significantly (P<0.05) except the 8th day (P>0.05).2、The level of ALT、AST: compared with the normal control group, ALT and AST raised without significant difference (P>0.05) but nearly physiological limit value.3、The level of BUN: compared with the normal control group, BUN raised without significant difference (P>0.05)Experiment 3:The exhausted swimming time: during the recovery period, the exhausted swimming time of the model group from the 1st to the 3rd day indicated a significant difference (P<0.05) except the 4th day (P>0.05) compared with the time before modeling.Conclusion:On the condition of indoor temperature (18~24℃), humidity (65±5%), light-controlled environment (light and dark, 12h alternation), the method, rats were forced swimming in a 120em×60em×60cm swimming pool filled with water (24±2℃) to a height of 30cm, 2h/d for 12 consecutive days, might establish a rat model of sub-health.ChapterⅡResearch on the effect of weikang granule on sub-health-resistanceObjective: To observed the effect of weikang granule on the exhausted swimming time、BUN and liver of the rat model of sub-health fatigue and discussed the mechanism and theory of sub-health.Method: 18 rats were divided into three groups: the normal control group (C), the model group (M) and the weikang granule group (WK). The C group was kept fasting and water deprived in cage when the M group and the WK group were modeling. Rats of the M group and the WK group kept forced-swimming 2h/d for two consecutive weeks in a 120cm×60cm×60cm swimming pool, 30cm deep and water temperature (24±2℃). The WK group rat received daily gavages for 2 weeks with lixivium of Weikang granule (0.14g/ml, 2.08g/kg wt). The exhausted swimming of the M group and WK group rat was recorded at the 12th day. All rats were executed and examined ALT、AST、BUN at the 14th day. Liver was taken pathological examination.Results:1、The exhausted swimming time: compared with the WK group, the exhausted swimming time of the M group decreased significantly at the 12th day (P<0.05).2、The level of ALT、AST: at the 14th day compared with the C group, AST of the M group increased significantly (P<0.05) but ALT not (P>0.05). The weikang group indicated no significant difference of ALT、AST with the C group (P>0.05).3、The level of BUN: at the 14th day compared with the C group and the WK group, BUN of the M group increased significantly (P<0.05). The weikang group indicated no significant difference of BUN with the C group (P>0.05).4、The pathological examination: All rats indicated no obvious changes in liver at the 14th day.Conclusion:Weikang granule might have a good effect on anti-sub-health and defend injury of liver caused by fatigue.
