On the clinical treatment of cancer and immune system diseases,the use of largedoses of radiation or high dose of chemotherapy drugs in the body “tolerance” dose,some recipients SSCs within the testicular destruction, even exhausted, causingnon-obstructive azoospermia formation. With the development of modern medicine,pre-adolescent cancer patients in clinical cure rate has exceeded70%, but15%ofthese tumor cure patients need to face the problem of infertility caused bychemotherapy or radiotherapy in a very long period of time. However,non-obstructiveazoospermia patients,whose spermatogenic function block for lymphoma or leukemiabefore puberty,varicocele,the parotid borne orchitis and other diseases,the medicinehas no effective treatment method. LBP is the main active ingredient of wolfberrybiological activity. Modern pharmacological experiments show that LBP hasimmunomodulatory, anti-aging, hypoglycemic, hepatoprotective, anti-radiation,anti-fatigue,the protection of the reproductive system,anti-tumor,anti-hypoxia andother functions. In the protection of the reproductive system, studies have shownthat LBP has a protective effect on spermatogenic cell. This experiment intends tocreate high-dose chemotherapy in mice infertility model with busulfan, and give LBPtreatment, then using c-kit as an observed indicator to detect spermatogenicfunctional recovery, to research the influence of LBP on the mouse testis expressionof c-kit factor after high-dose chemotherapy, and to further explore a new method oftreatment of infertility caused by high-dose chemotherapy. Objective To explore the influence of lycium barbarum polysaccharides ontestis c-kit gene expression in mice after high-dose chemotherapy.Methods4to6-week-old male Kunming White (KM)50mice were randomlydivided into the chemotherapy+saline group(model group), the chemotherapy+50mg/kg d-1LBP group, chemotherapy+100mg/kg d-1LBP group, chemotherapy+200mg/kg d-1LBP group(three treatment groups) and the normal control group,(n=10). To observe the general condition of the animals, the detection of infertilitymodels by HE staining, and the effect of LBP on testis c-kit factor expression in eachexperimental group by using immunohistochemistry, quantitative-polymerase chainreaction(q-PCR) and Western-blot.Results The model group (chemotherapy+saline group) compared with thenormal control group, the expression of the c-kit factor decreased significantly(allP<0.05);compared with the negative control group,the expression of c-kit factor withthe LBP treatment of the three groups have significantly higher(all P<0.05); theexpression of c-kit factor pairwise comparisons between chemotherapy+100mg/kg d-1LBP group, chemotherapy+200mg/kg d-1LBP group and normal control group,the difference was not statistically significant(all P>0.05).Conclusion LBP can significantly improve the expression of the c-kit factor inmouse testis after high-dose chemotherapy,100mg/kg d-1may be the best startingdose of LBP on the mouse spermatogenic functional recovery after high-dosechemotherapy.
