Puerarin and its inhibition rate of skin aging mice on total antioxidant capacity and hydroxyl free radical

Abstract


To study the effect of puerariae and its main components puerarin to the skin of natural aging mouse on activities of T-AOC and inhibiting rate of hydroxyl radical (·OH), and to observe the organizational structure of mouse skin before and after changes in drug use, to detect the function of puerarin in delaying the role of natural aging in mouse skin.Material and method:15of female Kunming mice of4-month-old as the young control group (A),90of female Kunming mice of18-month-old as the old age group, and then were randomly divided into6groups, with every group having15Kunming mice of18-month-old:elderly control group (B), positive control group of older (C), old age gypenoside group (D), puerarin high dose(E), puerarin low dose(F). And A group and B group fill the stomach physiological saline (20ml/kg/d), C group fills the stomach Vitamin EC suspl30mg/kg/d, D group fills the stomach gypenoside suspl30mg/kg/d, E group、F group fill the stomach puerarin suspl30mg/kg/d、15mg/kg/d. Every day fills the stomach one time, The first31days animals were sacrificed and materials.Results:1. Compared with young control group(A), the activities of T-AOC in skin of organizational structure of other six old-group were significantly decreased than group A, with significant differences P-value is less than0.01, the inhibitory rate of·OH significantly decreased P-value is less than0.01.2. Compared with group B, the activities of T-AOC were increased significantly in skin of organizational structure of group C, D, E,Fand G(respectively was: p<0.01, p<0.01, p<0.01, p<0.01, p<0.01), and increased the inhibitory rate of·OH in skin of organizational structure (respectively was:p<0.01, p<0.01, p<0.01, p<0.01, p<0.01).3. Morphological observations indicate that:compared with the young control group (A), the structure changes of epidermal and dermal elderly control group (B) were in state of aging; but in the positive control group (C), high-dose group of Kuzune (D), lower-dose group of Kuzune (E), high-dose group of puerarin (F), lower-dose group of puerarin (G), the structure of epidermis and dermis were better than the elderly control group (B), had not yet achieved the state of the skin structure of young control group (A).Conclusion:1. The skin of natural aging mouse on the activities of T-AOC and the inhibitory rate of·OH were significantly decreased.2. For it can clean radical and increase the activities of antioxidant, Puerarin can abate the oxidative damage of the skin of natural aging mouse. It also can increase the activities of T-AOC and the inhibitory rate of·OH so that to make skin aging slowly.3. From the histological observation:Compared with the young control group, skin the natural aging mice showed aging performance, Pueraria lobata and puerarin could lighten the morpholiqi change of aging skin.
